Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained.

Phosphorylation of specific tyrosine residues is the result of activation or stimulation of their respective protein tyrosine kinases. The phosphorylated proteins can be auto-phosphorylated kinases or certain cellular protein substrates. Tyrosine-phosphorylated proteins are involved in signal transduction and in the regulation of cell proliferation. Antibody to phosphotyrosine provides an excellent tool for the detection, characterization, and purification of phosphotyrosine containing proteins. This MAb shows no cross-reaction with other phosphoamino acids and is superb for multiple applications including staining of formalin/paraffin tissues.

This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia.

Recognises a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO). This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.

This antibody recognises both the free and protein-conjugated (either soluble or cell bound) form of biotin. The mAb is highly specific to biotin and shows no cross-reaction with other structurally related compounds. It has a very high affinity and is excellent for use in various amplification techniques. In some applications, localization of biotinylated probes with avidin produces unacceptably high background staining. Biotin antibody may be substituted to decrease this noise.
