Beschreibung:   The charged multivesicular body proteins, commonly designated CHMPs, belong to the vacuolar sorting protein family and function as chromatin-modifying proteins. CHMP1-6 are all components of ESCRT (endosomal sorting complex required for transport) I, II or III complexes. These complexes are crucial for sorting endosomal articles into multivesicular bodies (MVBs), and are also required for the formation of these bodies. CHMP2B, also known as CHMP2.5 or vacuolar protein-sorting-associated protein 2-2, is a 213 amino acid cytosolic protein. Widely expressed in brain, heart, skeletal muscle, small intestine, pancreas, lung, placenta and leukocytes, CHMP2B associates directly with CHMP2A and vps4 for the disassembly of the ESCRT-III complex. Defects in the gene encoding CHMP2B have been shown to cause chromosome 3-linked frontotemporal dementia (FTD3).

Beschreibung:   The alcohol dehydrogenase family of proteins metabolize a wide variety of substrates, including retinol, hydroxysteroids, ethanol, aliphatic alcohols and lipid peroxidation products. ADH5 (alcohol dehydrogenase 5 (class III)), also known as FDH (formaldehyde dehydrogenase), ADHX, ADH-3 or GSNOR, is a 374 amino acid cytoplasmic protein that belongs to the class III subfamily of alcohol dehydrogenases. Expressed ubiquitously, ADH5 uses iron as a cofactor to catalytically oxidize both long-chain primary alcohols and S-hydroxymethyl-glutathione, a product formed spontaneously between formaldehyde and glutathione. ADH5 exists as a homodimer and, via its ability to oxidize S-hydroxymethyl-glutathione and, thus, eliminate formaldehyde, functions as an important component of cellular metabolism. Genetic variations in the gene encoding ADH5 may affect drug and alcohol dependence in humans.

Beschreibung:   Dipeptidyl peptidases (DPPs) mediate regulatory activity of their substrates and have been linked to a variety of diseases including type 2 diabetes, obesity and cancer. DPPs have post-proline dipeptidyl aminopeptidase activity, cleaving Xaa-Pro dipeptides from the N-termini of proteins. DPPs can bind specific voltage-gated potassium channels and alter their expression and biophysical properties and may also influence T cells. DPP proteins include DPRP1, DPRP2, DPP3, DPP7, DPP10, DPPX and CD26. DPP10 (dipeptidyl-peptidase 10), also known as DPRP3 (dipeptidyl peptidase IV-related protein 3), DPL2 or DPPY, is a non-functional dipeptidyl peptidase which can bind to the potassium channels KV4.1 and KV4.2. It is a single-pass type II membrane protein expressed in spinal cord, adrenal glands, pancreas and brain tissues and may act as a modulator for cell surface expression and activity of KV4.1 and KV4.2.

Beschreibung:   Apolipoprotein E, a main apoprotein of the chylomicron, binds to a specific receptor on liver cells and peripheral cells and is essential for the normal catabolism of triglyceride-rich lipoprotein constituents.ApoE exists in three major isoforms; E2, E3, and E4, which differ from one another by a single amino-acid substitution. Compared with E3 and E4, E2 exhibits the lowest receptor binding affinity. Defects in ApoE are a cause of hyperlipoproteinemia type III due to increased plasma cholesterol and triglycerides levels which are the consequence of impaired clearance of chylomicron and VLDL remnants.Summary: Chylomicron remnants and very low density lipoprotein (VLDL) remnants are rapidly removed from the circulation by receptor-mediated endocytosis in the liver. Apolipoprotein E, a main apoprotein of the chylomicron, binds to a specific receptor on liver cells and peripheral cells. ApoE is essential for the normal catabolism of triglyceride-rich lipoprotein constituents. The APOE gene is mapped to chromosome 19 in a cluster with APOC1 and APOC2. Defects in apolipoprotein E result in familial dysbetalipoproteinemia, or type III hyperlipoproteinemia (HLP III), in which increased plasma cholesterol and triglycerides are the consequence of impaired clearance of chylomicron and VLDL remnants. Beschreibung:   GALR3 a 368 and 370 amino acid protein in human and rat, respectively, belongs to a family of G protein-coupled receptors that bind the neuropeptide galanin, which is distributed throughout the central and peripheral nervous system, the pituitary gland, the gastrointestinal tract and in the endocrine and exocrine pancreas. GALR3 mRNA is widely distributed, but expressed at low abundance. In human, GALR3 mRNA is highly expressed in the hypothalamus, pituitary and testis, and is expressed to a lesser extent in adrenal gland and pancreas. Rat and human GALR3 co-express with potassium channel subunits GIRK1 and GIRK4. Like GALR1, GALR3 signaling pathways lead to the inhibition of adenylate cyclase and to the activation of potassium channels, which are linked to the regulation of neurotransmitter release. Binding of galanin to galanin receptors results in increased feeding, impaired learning, enhanced opiate analgesia and decreased opiate place preference.
