The leucine-rich (LRR) repeat is a 20-30 amino acid motif that forms a hydrophobic å/∫ horseshoe fold, allowing it to accommodate several leucine residues within a tightly packed core. All LRR repeats contain a variable segment and a highly conserved segment, the latter of which accounts for 11 or 12 residues of the entire LRR motif. FLRT1 (fibronectin leucine rich transmembrane protein 1) is a 646 amino acid single-pass type I membrane protein that contains one fibronectin type-III domain and ten LRR repeats. Expressed in kidney and brain, FLRT1 is thought to play a role in cell adhesion and receptor signaling. FLRT1 shares similarity with FLRT2 and FLRT3 and is subject to post-translational N-glycosylation. The gene encoding FLRT1 maps to human chromosome 11, which houses over 1,400 genes and comprises nearly 4% of the human genome.

The Dumpy-30 (DPY-30) protein was first described in C. elegans, in which it is involved in dosage compensation of sex chromosomes. Conserved from yeast to humans, the DPY-30 family is involved in gene expression and chromatin modification, specifically histone methylation. As a member of the DPY-30 family, DYDC1 (DPY30 domain-containing protein 1), also known as DPY30D1 and RSD9, is a 177 amino acid protein that binds to Endophilin III and plays a crucial role during acrosome biogenesis. DYDC1 is specifically expressed in brain and testis and accumulates in the acrosome area during spermatogenesis. Knockdown of DYDC1 mRNA results in disruption of acrosome formation and spermatid differentiation.

TRIM36 (tripartite motif-containing 36), also known as RNF98 (RING finger protein 98), HAPRIN (haploid germ cell-specific RBCC protein) or RBCC728, is a 728 amino acid protein that belongs to the TRIM/RBCC (Ring finger, B box, coiled-coil) family. Predominantly expressed in prostate, testis and brain with weak expression in heart, kidney and lung, TRIM36 contains two B box-type zinc fingers, a SPRY domain, a coiled-coil domain, a fibronectin type-III domain and a RING-type zinc finger; a motif that has zinc-chelating activity and is involved in mediating protein-protein and protein-DNA interactions. Localizing to the cytoplasm and the acrosomal region of germ cells and mature sperm, TRIM36 is believed to play a role in the acrosome reaction and fertilization. In addition, TRIM36 is overexpressed in prostate cancer, suggesting a possible role for TRIM36 in prostate tumorigenesis.

This gene encodes the acidic form of complement factor 4, part of the classical activation pathway. The protein is expressed as a single chain precursor which is proteolytically cleaved into a trimer of alpha, beta, and gamma chains prior to secretion. The trimer provides a surface for interaction between the antigen-antibody complex and other complement components. The alpha chain may be cleaved to release C4 anaphylatoxin, a mediator of local inflammation. Deficiency of this protein is associated with systemic lupus erythematosus and type I diabetes mellitus. This gene localizes to the major histocompatibility complex (MHC) class III region on chromosome 6. Varying haplotypes of this gene cluster exist, such that individuals may have 1, 2, or 3 copies of this gene. Two transcript variants encoding different isoforms have been found for this gene. CP2 family gene products are reported to be involved in a-globin gene expression. Mouse CP2c (also known as a-CP2/CP2/LSF/UBP-1) is a homologue of human LBP-1c with 96% amino acid sequence identity. CP2c was initially identified as an activator of the mouse a-globin gene, which binds to the consensus DNA-binding sequence CNRG-N6-CNR(G/C) within a promoter element overlapping the CCAAT box. The transcriptional activity of CP2c was increased during induced differentiation of MEL cells and associated globin gene transcription. CP2c is also likely to be involved in regulation of non-globin erythroid-specific genes. It was reported that congenital erythropoietic porphyria was caused by the mutations in GATA-1 and CP2c binding sites within the promoter of the uroporphyrinogen III synthase gene, the fourth enzyme in the heme biosynthetic pathway.

This antibody recognizes a cell surface glycoprotein of 95/115/135 kDa (depending upon the extent of glycosylation), identified as CD43 (Workshop III). 70-90% of T-cell lymphomas and from 22-37% of B-cell lymphomas express CD43. No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with anti-CD20, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with anti-CD20 and anti-CD43 argues against a reactive process and favors a diagnosis of lymphoma.
