Potassium channels represent the most complex class of voltage-gated ion channels from both functional and structural standpoints. Their diverse functions include regulating neurotransmitter release, heart rate, insulin secretion, neuronal excitability, epithelial electrolyte transport, smooth muscle contraction, and cell volume. Four sequence-related potassium channel genes - shaker, shaw, shab, and shal - have been identified in Drosophila, and each has been shown to have human homolog(s). This gene encodes a member of the potassium channel, voltage-gated, shaker-related subfamily. This member contains six membrane-spanning domains with a shaker-type repeat in the fourth segment. It belongs to the delayed rectifier class, members of which allow nerve cells to efficiently repolarize following an action potential. It plays an essential role in T-cell proliferation and activation. This gene appears to be intronless and it is clustered together with KCNA2 and KCNA10 genes on chromosome 1. The activity of this receptor is mediated by G-proteins that inhibit adenylyl cyclase activity, stimulates phospholipase A2, activates potassium channels, inactivates voltage-dependent calcium-channels and modulates inositol phospholipids hydrolysis. Plays a critical role in the fine-tuning of inhibitory synaptic transmission. Pre-synaptic GABA-B-R inhibit neurotransmitter release by down-regulating high-voltage activated calcium channels, whereas postsynaptic GABA-B-R decrease neuronal excitability by activating a prominent inwardly rectifying potassium (Kir) conductance that underlies the late inhibitory postsynaptic potentials. Not only implicated in synaptic inhibition but also in hippocampal long-term potentiation, slow wave sleep, muscle relaxation and antinociception.

The KIR family of potassium channels possess a greater tendency to allow potassium to flow into the cell rather than out of it. Kir4.1, also known as Kir1.2, is highly expressed in brain including glial cells, astrocytes and cortical neurons. Kir4.1 is also expressed in myelin-synthesizing oligodendrocytes and is crucial to myelination in the developing nervous system. The gene encoding human Kir4.1 maps to chromosome 1. Kir4.2, also known as Kir1.3, is expressed in kidney, lung, heart, thymus and thyroid during development. The gene encoding human Kir4.2 maps to chromosome 21 in the Down syndrome chromosome region 1, and Kir4.2 may play a role in the pathogenesis of Down’s syndrome. Kir 5.1 forms functional channels only by coexpression with either Kir4.1 or Kir4.2 in the kidney and pancreas. The gene encoding human Kir5.1 maps to chromosome 17.
