EYA3 is a 573 amino acid protein that localizes to both the nucleus and the cytoplasm and is one of several mammalian homologs of the Drosophila Eya (eyes absent) protein. Existing as two alternatively spliced isoforms, EYA3 possesses magnesium-catalyzed phosphatase activity and is thought to play a role in transcriptional regulation during organogenesis. Specifically, EYA3 interacts with proteins such as Six1 and, via this interaction, functions to activate the expression of genes that are involved in cellular proliferation and organ development. Upon DNA damage, EYA3 may be phosphorylated by ATM or ATR. The gene encoding EYA3 maps to chromosome 1, which spans about 260 million base pairs and comprises nearly 8% of the human genome.

It is possible that the p41 subunit is involved in assembling and maintaining the structure of the Arp2/3 complex. Multiple versions of the p41 subunit may adapt the functions of the complex to different cell types or developmental stages. With 181 million base pairs encoding around 1,000 genes, chromosome 5 is about 6% of human genomic DNA. It is associated with Cockayne syndrome through the ERCC8 gene and familial adenomatous polyposis through the adenomatous polyposis coli (APC) tumor suppressor gene. Treacher Collins syndrome is also chromosome 5 associated and is caused by insertions or deletions within the TCOF1 gene. Deletion of the p arm of chromosome 5 leads to Cri du chat syndrome. Deletion of 5q or chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.

The VPS10 domain name comes from the yeast carboxypeptidase Y sorting receptor Vps10 protein. Members of this gene family are large with many exons but the CDS lengths are usually less than 3700 nt. Very large introns typically separate the exons encoding the VPS10 domain; the remaining exons are separated by much smaller-sized introns. These genes are strongly expressed in the central nervous system. Two of the five family members (sortilin and sortilin-related receptor) are synthesised as preproproteins; it is not yet known if this encoded protein is also a preproprotein.
