The retinal pigment epithelium (RPE) is a monolayer simple epithelium in proximity to the outer surface of the retinal photoreceptor cells. Retinal pigment epithelium-specific protein (RPE65) is a 65 kDa protein belonging to the β-carotene dioxygenase family. This protein is important in 11-cis retinal production as well as in visual pigment regeneration. RPE65 is attached to the membrane by a lipid anchor when palmitoylated (membrane form) and soluble when unpalmitoylated. The soluble form of the protein binds vitamin A. Defects in RPE65 causes autosomal dominant retinitis pigmentosa and/or Leber congenital amaurosis type 2.

HSP56 is a cis-trans prolyl isomerase belonging to the immunophilin protein family. The human HSP 56 gene (FKBP4) has multiple polyadenylation sites and the HSP 56 protein can undergo phosphorylation. HSP 56 influences immunoregulatory gene expression in lymphocytes, protein folding and trafficking. It can serve as a co-chaperone for steroid hormone nuclear receptors to govern appropriate hormone action in target tissues. The protein can associate with phytanoyl-CoA alpha-hydroxylase (PHYH) and with HSP90 through a series of tetratricopeptide repeat (TPR) domains. HSP 56 is a TRPC ion channel accessory protein that modulates channel activation following receptor stimulation.

GM130, a cis-Golgi matrix protein, interacts specifically with p115 and provides a membrane docking site. Both GM130 and p115 are involved in vesicle tethering to Golgi membranes. The protein p115 also binds p400, alternatively called giantin. Giantin, the majority of whose mass projects into the cytoplasm, is involved in the docking of COPI vesicles via p115 to the Golgi membrane. Giantin, which also is known as macrogolgin or Golgi complex-associated protein, is involved in cross-bridge formation in the Golgi complex. Contactin 2 is a neuronal cell adhesion molecule (CAM) that influences the formation of axon connections in the developing nervous system. Contactin 2 is a member of the immunoglobulin superfamily (IgSF) and contains a glycosylphosphatidylinositol-anchor, six immunogobulin (Ig)-like and four Fibronectin type III (FNIII)-like domains. Contactin 2 is expressed predominantly during neural development on the cell membrane of axons in nerve fiber tracts in order to guide commissural axons without promoting their growth. Contactin 2 binds with NgCAM in the plane of the same membrane (cis-binding). The Contactin 2 heterophilic (Contactin 2/NgCAM and Contactin 2/NrCAM) binding sites are localized to the first four Ig domains. The Contactin 2 homophilic (Contactin 2/Contactin 2) binding site is localized to the FNIII domain.
