SMYD3 is a gene that is over-expressed in the majority of colorectal carcinomas and hepatocellular carcinomas. SMYD3 forms a complex with RNA polymerase II through an interaction with the RNA helicase HELZ and transactivates a set of genes that included oncogenes, homeobox genes and genes associated with cell-cycle regulation. SMYD3 binds to a motif, 5'-CCCTCC-3', present in the promoter region of downstream genes such as Nkx2.8. The SET domain of SMYD3 shows histone H3-lysine 4 (H3-K4)-specific methyltransferase activity, which is enhanced in the presence of the heat-shock protein HSP90A.

NaBC1 is a protein found amplified in most breast carcinoma forms. It is expressed primarily as a cytoplasmic, detergent-stable homodimer that has a tendency to interact with DYNLL1 (PIN) and DYNLL2. Breast tumor lines that exhibit 20q13.2 gene amplification express much higher levels of the protein as compared to the levels found in other breast cancer lines that do not overexpress the NaBC1 mRNA. However, this upregulation does not affect growth rate or anchoring abilities of a cell, indicating the oncogenic properties of NaBC1 differ from that of other oncogenes.

Perilipin is an intracellular neutral lipid droplet protein that is hormonally regulated. This protein is localized exclusively to the surface of lipid droplets. In response to lypotic stimuli, perilipin is phosphorylated by protein kinase A. Once activated, perilipin has inhibitory affects upon hormone-sensitive lipase (HSL), a protein that mediates the hydrolysis of triacylglycerol, the major form of stored energy in the body. Perilipin expression is limited to adipocytes and steroidogenic cells. There are currently two known isoforms, Perilipin A and B. Both of these proteins are encoded by a single-copy gene and are the result of differential splicing events.
