Protein kinases (PKs) represent a well studied but most diverse protein superfamily. The covalent, reversible linkage of phosphate to serine, threonine, and tyrosine residues of substrate proteins by protein kinases is probably ubiquitous cellular mechanism for regulation of physiological processes. It is known to us that most signaling pathways impinge at some point on protein kinases. Here we report a human putative receptor protein kinase cDNA STYK1. The STYK1 cDNA is 2749 base pairs in length and contains an open reading frame encoding 422 amino acids. The STYK1 gene is mapped to human chromosome 12p13 and 11 exons were found. RT-PCR showed that STYK1 is widely expressed in human tissues.

Heat Shock Protein beta -11 (HSPB11) is a stress-responsive protein that is required to deal with proteotoxic stresses. HSPB11 is composed of an IFT complex B composed of IFT88, IFT57, TRAF3IP1, IFT52, IFT27, HSPB11 and IFT20 and is detected in placenta. HSPB11 has beeb shown to form oligomeric complexes and prevent the aggregation of in vitro denaturated aldolase and glyceraldehyde-3-phosphate dehydrogenase in accordance with the chaperone model of HSPB1 and HSPB5. HSPB11 overexpression protected against etoposide-induced cell death that correlated with a decreased release of mitochondrial Cytochrome C into the cytosol. Inhibition of HSP90 function completely abrogated the protective effect of HSPB11. This data suggests that at least in the case of HSPB11, interaction with other chaperone machines besides HSPA1A may contribute to functional specificity and cellular functioning.

The antibody is produced by hyperimmunizing goats with the enzyme beta-galactosidase from Escherichia coli. The IgG fraction is prepared by delipidation, ammonium sulfate fractionation, and anion exchange chromatography, and is further purified by solid-phase absorption to remove antibodies to non-beta-galactosidase E. coli proteins. The product is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, adjusted to standard titer, filtered through a 0.22 μm filter, and vialed. No preservative is added.

Slc37A2 Antibody: The Slc37 gene family consists of four members which have 10-12 predicted transmembrane domains and belong to the sugar-phosphate transporter superfamily. Recent experiments have demonstrated that Slc37A2 is a macrophage-specific gene selectively expressed in obese white adipose tissue. Its transcript is also expressed in spleen and thymus and the cell lines RAW264.7 and THP-1. Compared to wild-type mice, Slc37A2 mRNA is expressed at nine-fold higher levels in abdominal epididymal fat deposits of ob/ob mice, suggesting that it may be required to limit the contribution of macrophages to the inflammatory state present in obesity. Slc37A2 protein is post-translationally modified with the addition of N-linked glycans and migrates as a heterogeneous species of 50-75kDa.

GLS2 Antibody: Phosphate-activated glutaminase, also known as Glutaminase 2 (GLS2), was initially isolated from rat liver, although it has been shown to be expressed in other tissues. Like the functionally similar, larger kidney glutaminase, GLS2 catalyzes the hydrolysis of glutamine to stoichiometric amounts of glutamate and ammonia. Expression of GLS2 is increased by p53 under both stressed and nonstressed conditions, resulting in increased levels of glutamate and alpha-ketoglutarate, which in turn results in enhanced mitochondrial respiration and ATP generation. GLS2 also regulates antioxidant defense function in cells by increasing reduced glutathione levels and decreasing ROS-levels, suggesting that GLS2 acts as a mediator of p53's role in antioxidant defense in addition to its role in energy metabolism.

Alkaline Phosphatase, Tissue-Nonspecific Isozyme (ALPL) is a cell membrane protein which belongs to the alkaline phosphatase family. There are at least four distinct but related alkaline phosphatases in humans: intestinal AP (IAP), placental AP(PLAP), germ cell AP (GCAP) and their genes are clustered on chromosome 2, tissue-nonspecific isozyme (TNAP) which gene is located on chromosome 1. Alkaline phosphatases (APs) are dimeric enzymes, it catalyse the hydrolysis of phosphomonoesters with release of inorganic phosphate. The native ALPL is a glycosylated homodimer attached to the membrane through a GPI-anchor. This isozyme may play a role in skeletal mineralisation. Mutations in ALPL gene have been linked directly to different forms of hypophosphatasia,characterised by poorly mineralised cartilage and bones, and this disorder can vary depending on the specific mutation since this determines age of onset and severity of symptoms.

GPAT1 Antibody: Glycerol-3-phosphate acyltransferase 1 (GPAT1), one of four known GPAT isoforms, is located on the mitochondrial outer membrane, allowing reciprocal regulation with carnitine palmitoyltransferase-1. It is thought to be critical for the development of hepatic steatosis; steatosis triggered by GPAT1 overexpression leads to hepatic and possibly peripheral insulin resistance. GPAT1 is transcriptionally upregulated by insulin and sterol regulatory element binding protein (SREBP-1) and downregulated by AMP-activated protein kinase. Mice deficient in GPAT1 exhibit decreased triacylglycerol (TAG) in cardiomyocytes even in high-fat diets, suggesting that GPAT1 contributes significantly to TAG accumulation in heart tissue during lipogenic or high fat diets. At least two isoforms of GPAT1 are known to exist.

The amine-reactive portion of SPDP reagents is the N-hydroxysuccinimide (NHS) ester. Reactions are most commonly performed in phosphate, carbonate/bicarbonate, or borate buffers at pH 7-8. Other buffers can be used provided they do not contain primary amines (or thiols or disulfide reducing reagents – see discussion of the sulfhydryl reactivity). The rate of reaction and degradation by hydrolysis increases with increasing pH; for example, the half-life of the NHS ester is several hours at pH 7 and less than 10 minutes at pH 9. NHS-ester reagents like SPDP and LC-SPDP have limited aqueous solubility and must be dissolved in organic solvent before adding them to a reaction mixture. Sulfo-NHS-ester reagents like Sulfo-LC-SPDP are water-soluble and can be added directly to aqueous reaction mixtures.

ALAS-1 (EC 2.3.1.37) is a nuclear-encoded mitochondrial matrix enzymecatalyzing the condensation of glycine with succinyl-CoA to form delta-amino levulinate, CO2 and CoA. It regulates the first and rate-limiting step of heme biosynthetic pathway. It is one of the two isoforms of ALAS and is a pyridoxal-5 phosphate dependant housekeeping enzyme. It is ubiquitously expressed and is responsible of providing heme for cytochromes and other hemoproteins. ALAS1 in liver undergoes negative feedback regulation by heme. Transcription of ALAS1 gene is stimulated by cAMP and respiratory uncoupling whereas Phorbol esters and insulin repress ALAS1 gene expression. ALAS1 is suggested to be involved in reciprocal regulation of Heme biosynthesis and circadian clock suggesting a potential target for treatment of circadian disorders. ALAS1 gene expression is down regulated in Acute Liver failure resulting in altered heme metabolism and liver function.
