Beschreibung:   Interleukins (ILs) are a large group of cytokines that are produced mainly by leukocytes, although some are produced by certain phagocytes and auxiliary cells. ILs have a variety of functions, but most function to direct other immune cells to divide and differentiate. Each IL acts on a specific, limited group of cells through a receptor specific for that IL. Human IL10 is a non glycosylated polypeptide consisting of 178 amino acids. There is 73% homology between the human and mouse IL10 proteins, however, the human IL10 acts on both human and mouse target cells, while the mouse IL10 has species specific activity. The cellular sources of IL10 are CD4+ T cells and T cell clones, thymocytes, B cells and B cell lymphomas, macrophages, mast cell lines and keratinocytes. IL10 will stimulate the growth of stem cells, mast cells and thymocytes. IL10 enhances cytotoxic T cell development, and costimulates B cell differentiation and immunoglobulin secretion. IL10 inhibits cytokine production by macrophages and suppresses macrophage class II MHC expression. The human IL10 gene is on human chromosome 1.

Beschreibung:   Human Recombinant Angiotensin-Converting Enzyme 2 (ACE2) Protein, aa18-740, is expressed in HEK293 cells. ACE2 activity results in vasodilation by degrading vasoconstrictor angiotensin II to produce vasodilator angiotensin. Decreased expression of this enzyme is associated with cardiovascular disease and possibly also glomerular injury in diabetic nephropathy. ACE2 contains a peptidase domain at the N-terminus and a single transmembrane helix. ACE2 is also the primary human receptor for the surface spike glycoprotein of SARS-CoV-2 and SARS-CoV. This receptor is expressed in human lung and small intestine epithelia as well as the heart, kidney, and testes. The peptidase domain of ACE2 binds to the protein-binding domain of the S1 protein subunit of the spike protein; this binding results in a cleavage site becoming exposed. Cleavage, or ‘protein priming’, is done by TMPRSS2, a cellular serine protease, and by cathepsins. This cleavage activates the S2 subunit of the spike protein, allowing the fusion of the viral membrane to the host cell membrane. Renin activates the renin-angiotensin system by cleaving angiotensinogen, produced by the liver, to yield angiotensin I, which is further converted into angiotensin II by ACE, the angiotensin-converting enzyme primarily within the capillaries of the lungs. Renin is secreted from kidney cells, which are activated via signaling from the macula densa, which responds to the rate of fluid flow through the distal tubule, by decreases in renal perfusion pressure (through stretch receptors in the vascular wall), and by sympathetic nervous stimulation, mainly through beta-1 receptor activation. Renin can bind to ATP6AP2, which results in a fourfold increase in the conversion of angiotensinogen to angiotensin I over that shown by soluble renin. In addition, renin binding results in phosphorylation of serine and tyrosine residues of ATP6AP2. The level of renin mRNA appears to be modulated by the binding of HADHB, HuR and CP1 to a regulatory region in the 3' UTR. An over-active renin-angiotension system leads to vasoconstriction and retention of sodium and water. These effects lead to hypertension. The KRT8 gene is a member of the type II keratin family clustered on the long arm of chromosome 12. Cytokeratin 8 participates in cellular differentiation and signal transduction, protects against apoptosis, stress and injury, and helps maintain cellular structural integrity. It is primarily found in the non-squamous epithelia and is present in majority of adenocarcinomas and ductal carcinomas. It is absent in squamous cell carcinomas. Specific combinations of cytokeratins are associated with certain epithelial cells, and therefore useful in the characterization of poorly differentiated carcinoma. Hepatocellular carcinomas are defined by the use of antibody that recognises only cytokeratin 8 and 18. Keratin 8 exists on several types of normal and neoplastic epithelia, including many ductal and glandular epithelia such as colon, stomach, small intestine, trachea, and esophagus as well as in transitional epithelium. Antibody to Cytokeratin 8 does not react with skeletal muscle or nerve cells. Epithelioid sarcoma, chordoma, and adamantinoma show strong positivity corresponding to that of simple epithelia (with antibodies against Keratin 8, 18 and 19). Reportedly, Cytokeratin 8 antibody is useful for the differentiation of lobular (ring-like, perinuclear) from ductal (peripheral-predominant) carcinoma of the breast.

The receptor for IL-22 (IL-22R, also termed CRF2-9 and IL-TIF-R1 chain) is a new member of the class II cytokine receptor family. IL-22R forms a complex with IL-10 receptor beta chain and mediates IL-22 signaling. IL-22 and its receptor activate JAK-STAT signaling pathway. A soluble form of IL-22 receptor, also termed IL-22 binding protein (IL-22BP) and IL-22 receptor-alpha 2 (IL-22RA2), was identified very recently.
