HIC-2 is also known as Hic-3, HIC1-related gene on chromosome 22 or Zinc finger and BTB domain-containing protein 30, and is a 615 amino acid protein that is expressed as two isoforms produced by alternative splicing. HIC-2 is highly expressed in cerebellum and is localised to the nucleus in cells. HIC-2 contains a short amino acid sequence that is thought to interact with CtBP, a transcriptional repressor. The gene sequence associated with HIC-2 is thought to be a target for miRNAs (microRNAs) which are expressed in many cancers, suggesting that HIC-2 could possess tumor suppressor capabilities.

The Src homology 3 (SH3) region is a small protein domain of approximately 60 amino acids present in a large group of proteins. In general, it exists in association with catalytic domains, as in the nonreceptor protein-tyrosine kinases and phospholipase C-? within structural proteins, such as spectrin or Myosin; and in small adapter proteins, such as Crk and GRB2. SH3 domains are often accompanied by SH2 domains of 100 amino acids, which bind to tyrosine-phosphorylated regions of target proteins, frequently linking activated growth factors to putative signal transduction proteins. The functions of SH3 domains are not as well defined. Deletion or mutation of SH3 domains generally activate the transforming potential of nonreceptor tyrosine kinases, suggesting that SH3 mediates negative regulation of an intrinsic transforming activity. 3BP1 has been identified as a protein with a high affinity proline-rich binding site for the SH3 domain of c-Abl p120. A similar putative adapter protein, designated Slap, for Src-like adapter protein, has been cloned. Slap contains a single SH2 and SH3 domain that exhibits homology with those from members of the Src kinase family. The N- and C-termini, however, are unique.

The G protein-coupled receptor TGR5 is a 330-amino acid protein that is almost universally expressed in human tissues including heart, skeletal muscle, spleen, kidney, liver, small intestine, placenta, and leukocytes, but not in brain, colon (without mucosa), thymus, or lung. TGR5 is sensitive to bile acids and responds through a significant mechanism that coordinates energy homeostasis. Bile acids activate mitogen-activated protein (MAP) kinase pathways, specifically induce TGR5 internalization, promote an increase of guanosine 5'-O-3-thio-triphosphate binding in membrane fractions, and cause rapid intracellular cAMP production. Bile acids also provoke TGR5 to suppress macrophage functions. TGR5-controlled signaling pathways may be good candidates for drug targets to treat common metabolic diseases, such as obesity, type II diabetes, hyperlipidemia, and atherosclerosis.
