Required for axonemal recruitment of CCDC39.Involvement in disease:Defects in CCDC40 are the cause of primary ciliary dyskinesia type 15 (CILD15) . A disorder characterised by abnormalities of motile cilia. Respiratory infections leading to chronic inflammation and bronchiectasis are recurrent, due to defects in the respiratory cilia; reduced fertility is often observed in male patients due to abnormalities of sperm tails. Half of the patients exhibit randomisation of left-right body asymmetry and situs inversus, due to dysfunction of monocilia at the embryonic node. Primary ciliary dyskinesia associated with situs inversus is referred to as Kartagener syndrome.

Modulation of the chromatin structure plays an important role in the regulation of transcription in eukaryotes. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

RORET, also known as RING finger protein 15 (RNF15) or zinc finger protein RoRet, is a 465 amino acid member of the TRIM family, also known as the RING-B-box coiled-coil (RBCC) family. Members of the RBCC family have an N-terminal RING finger, followed by one or two zinc-binding domains (B-box domains), a leucine coiled-coil region and a variable C-terminal domain. Found in all eukaryotes, members of the RBCC family typically function within a larger protein complex and possess ubiquitin-protein isopeptide ligase activity.The protein encoded by this gene is a member of the tripartite motif (TRIM) family. The TRIM motif includes three zinc-binding domains, a RING, a B-box type 1 and a B-box type 2, and a coiled-coil region. The function of this protein has not been identified.

RAS superfamily comprises around 50 related genes encoding GTP-binding domain (G-proteins) involved in signal transduction. The main genes are HRAS, NRAS and KRAS. Ras proteins are membrane-bound GTPases. The inactive form is GDP-bound. They are activated by ligand-binding receptor tyrosine kinases such as EGFR, PDGFR, colony-stimulating factor and fibroblast growth factor. These kinases transiently convert RAS-GDP to RAS-GTP, the active form of RAS. Single amino acid substitutions can activate RAS making it highly oncogenic. Such mutations generally reduce the GTPase activity of RAS, prolonging it in its active GTP-bound form. The consequence of this is sustained activation of the RAF1-MAPK signalling pathway. RAS mutations are found in 10-15% of tumours. A high incidence of RAS mutations is found in pancreatic cancers.

Interleukin-15 (IL-15) has a broad spectrum of biological activities. It is crucial for the development, proliferation, survival and differentiation of multiple cells from both innate and adaptive immune systems. IL-15 up-regulation has a central role in the development of several autoimmune or chronic inflammatory disorders. Targeting IL-15 or its receptor may have a valuable impact on the treatment of immune-mediated diseases. IL-15 participates in the development of important immune antitumour mechanisms. It activates CD8(+) T cells, natural killer (NK) cells, NK T cells, and can promote the formation of antitumour antibodies. IL-15 can also protect T effector cells from the action of T regulatory cells and reverse tolerance to tumour-associated antigens. In pre-clinical studies IL-15 has been found to demonstrate potentiated antitumour effects following pre-association with IL-15Ralpha, or when used in combination with chemotherapy, adoptive therapy, monoclonal antibodies, and tumour vaccines.
Application: Useful for immunofluorescent staining and flow cytometric analysis to identify and enumerate IL-15Ralpha expressing cells within mixed cell populations.
