APC7 Antibody: Cell cycle regulated protein ubiquitination and degradation within subcellular domains is thought to be essential for the normal progression of mitosis. APC7 is a highly conserved component of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. APC/C is responsible for degrading anaphase inhibitors, mitotic cyclins, and spindle-associated proteins ensuring that events of mitosis take place in proper sequence. The individual APC/C components mRNA and protein levels are expressed at approximately the same levels in most tissues and cell lines, suggesting that they perform their functions as part of a complex. APC7 is required for proper protein ubiquitination function of APC/C and for the interaction of APC/C with various transcription coactivators.

LIG4 encodes a DNA ligase that joins single-strand breaks in a double-stranded polydeoxynucleotide in an ATP-dependent reaction. This protein is essential for V (D)J recombination and DNA double-strand break (DSB) repair through nonhomologous end joining (NHEJ). This protein forms a complex with the X-ray repair cross complementing protein 4 (XRCC4), and further interacts with the DNA-dependent protein kinase (DNA-PK). Both XRCC4 and DNA-PK are known to be required for NHEJ. The crystal structure of the complex formed by this protein and XRCC4 has been resolved. Defects in this gene are the cause of LIG4 syndrome.

XRCC5 encodes the 80-kilodalton subunit of the Ku heterodimer protein which is also known as ATP-dependant DNA helicase II or DNA repair protein XRCC5. Ku is the DNA-binding component of the DNA-dependent protein kinase, and it functions together with the DNA ligase IV-XRCC4 complex in the repair of DNA double-strand break by non-homologous end joining and the completion of V (D)J recombination events. This gene functionally complements Chinese hamster xrs-6, a mutant defective in DNA double-strand break repair and in ability to undergo V (D)J recombination. A rare microsatellite polymorphism in this gene is associated with cancer in patients of varying radiosensitivity.

AIMP2 was initially identified as a part of an aminoacyl-tRNA synthesase complex. It was later discovered to be a cofactor and substrate of Parkin, a Ring-type E3 ubiquitin ligase that is important for the survival of dopamine neurons in Parkinson’s disease; accumulation of AIMP2 in these cells lead to catecholaminergic cell death. AIMP2 can also bind to TRAF2, a key player in the TNF-alpha signaling pathway, causing the ubiquitination of TRAF2 by cIAP1, leading to TNF-alpha-dependent apoptosis. Finally, AIMP2 has been suggested to function as a tumor suppressor.

The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. CHFR is an E3 ubiquitin-protein ligase required to transiently arrest cells in early prophase when they are exposed to microtubule poisons. It acts in early prophase before chromosome condensation, when the centrosome moves apart from each other along the periphery of the nucleus. CHFR probably promotes the formation of 'Lys-63'-linked polyubiquitin chains and functions with the specific ubiquitin-conjugating UBC13-MMS2 (UBE2N-UBE2V2) heterodimer. Substrates that are polyubiquitinated at 'Lys-63' are usually not targeted for degradation, but are rather involved in signaling cellular stress. This suggests that it may be involved in signaling the presence of mitotic stress caused by microtubule poisons.

Cbl-c is also known as signal transduction protein CBL-C, SH3-binding protein CBL-C, CBL-3, and RING finger protein 57. Cbl proteins are a family of ubiquitin protein ligases (E3s) that negatively regulate signaling by targeting activated tyrosine kinases for degradation. Cbl-c (a.k.a. Cbl-3) is the most recently cloned member of the Cbl proteins and is expressed only in epithelial cells (the other Cbl proteins are ubiquitously expressed). Cbl-c, like the other mammalian Cbl proteins, can ubiquitinate the activated EGFR and target it for degradation. Cbl-c knock out mice show no obvious phenotype. Thus, the physiological role of Cbl-c is not known.

CBX4 Antibody: Polycomb group (PcG) proteins are chromatin-associated proteins which are important for embryonic and adult stem cell self-renewal and maintenance. At least two distinct human PcG complexes have been identified. Polycomb repressive complex I (PRC1) contains CBX4/Pc2, HPH1 and RING domain-containing proteins (RING1, RING2 and BMI1). CBX4 (Chromobox protein homolog 4 or PC2), which functions as a transcriptional suppressor, is a candidate of KyoT2-binding proteins. It is a E3 SUMO-protein ligase which facilitates SUMO1 conjugation. Depletion of CBX4 results in decreased cellular resistance to ionizing radiation. CBX4 is directly involved in the cellular response to DNA damage.

The encoded protein is a nuclear phosphoprotein that binds and inhibits transactivation by tumor protein p53, as part of an autoregulatory negative feedback loop. MRE11 Antibody: MRE11 is involved in the repair of DNA double strand breaks as part of a complex that includes the Rad50 and NBS1 protein and is thought to act in the same pathway as the A-T mutated (ATM) protein. By itself, the protein has 3' to 5' exonuclease activity and endonuclease activity. The protein forms a complex with the RAD50 homolog; this complex is required for non-homologous joining of DNA ends and possesses increased single-stranded DNA endonuclease and 3' to 5' exonuclease activities. In conjunction with a DNA ligase, this protein promotes the joining of noncomplementary ends in vitro using short homologies near the ends of the DNA fragments. Mutations in this protein result in a novel ataxia telangiectasia-like disorder (ATLD). Unlike the ATM protein, MRE11 is necessary proper mammalian development.
