Phosphatidylethanolamine (PtdEtn) is a major membrane phospholipid which serves to play a primary role in cell membrane structure and is also involved in cell division, cell signaling, activation, phagocytosis and autophagy. PCYT2 (Phosphorylethanolamine transferase), also known as Ethanolamine-phosphate cytidylyltransferase, is a 389 amino acid protein that catalyzes the formation of CDP-ethanolamine from ethanolamine. This product combined with diacylglycerol form phosphatidylethanolamine via the de novo Kennedy pathway. PCYT2 is expressed at highest levels in heart, liver and skeletal muscle. Elevated levels of MyoD, reduced content of Sp1 and a changed ratio of Sp1 to Sp3 all together stimulate upregulation of PCTY2 transcription during C2C12 muscle cell differentiation. Disruption of the PCYT2 gene in mice leads to death after embryo implantation, establishing the necessity of PCYT2 for murine development.

The protein encoded by this gene is a type I transmembrane protein that localizes to the endocytic pathway. This protein contains a RING zinc-finger motif and has been shown to possess E3 ubiquitin ligase activity. Expression of this gene in retrovirally transduced T cell hybridoma significantly inhibits activation-induced IL2 and IL4 cytokine production. Induced expression of this gene was observed in anergic CD4(+) T cells, which suggested a role in the induction of anergic phenotype. Other peptides, including chromostatin, beta-granin, WE-14 and GE-25, are also derived from the full-length protein. However, biological activities for these molecules have not been shown. DNA polymerase lambda (pol Kappa), also designated DNA polymerase Kappa or Pol Beta2, is a low-fidelity polymerase which plays a role in both spontaneous and DNA damage-induced mutagenesis. Encoded by the POLL gene, pol Kappa is a member of the DNA polymerase type-X family. Pol Kappa extends primer-terminal mispairs opposite nondamaged DNA templates, suggesting that it may assist in extending mismatched base pairs during normal DNA replication. In addition, pol Kappa may play a role in the mutagenic bypass of T-T dimers. Proliferating cell nuclear antigen (PCNA), a protein essential to DNA replication, interacts with pol Kappa and thus influences the ability of pol Kappa to synthesize DNA.

NF-1, also designated CTF, consists of a family of CCAAT box binding proteins that stimulate DNA replication and activate transcription. Analysis of human NF-1 messenger RNA has revealed two forms of the NF-1 protein arising from an alternate splicing of a single NF-1 gene. NF-1 binds its consensus DNA element as a homodimer via an amino-terminal DNA binding domain, and activates transcription through a putatively novel, proline-rich, carboxy terminal transactivation domain. The NF-1 protein has been shown to recognize and bind the adenovirus type 2 promoter and activate transcription of herpes simplex virus thymidine kinase genes. The NF-1 consensus element has been found in the upstream promoter region of myriad eukaryotic genes, including that of Ha-Ras, alpha-globin, HSP 70, GRP 78, Histone H1, myelin basic protein and in the Xenopus laevis vitellogenin gene promoter.

The P2X receptor family is comprised of ligand-gated ion channels that allow for the increased permeability of calcium into the cell in response to extracellular ATP. The seven P2X receptors, P2X1-P2X7, form either homomeric or heteromeric channels or both. They are characterized by intracellular amino- and carboxy-termini. P2X receptors are expressed in a wide variety of tissues, including neurons, prostate, bladder, pancreas, colon, testis and ovary. The major function of the P2X receptors is to mediate synaptic transmissions between neurons and to other tissues via the binding of extracellular ATP, which acts as a neurotransmitter. The P2X receptors may be involved in the onset of necrosis or apoptosis after prolonged exposure to high concentrations of extracellular ATP.

Localizing to nucleus, EDRF1 is involved in transcriptional activation of globin genes by regulating DNA-binding activity of GATA-1 transcription factor. EDRF1 may also play an important role in organ development and histological differentiation. EDRF1 exists as four alternatively spliced isoforms and is encoded by a gene mapping to human chromosome 10q26.13.

G protein-coupled receptor 17, GPR17, also known as uracil nucleotide/cysteinyl leukotriene receptor or P2Y-like receptor (P2YL), is a 367 amino acid member of the G-protein coupled receptor 1 family of proteins. While GPR17 is expressed in kidney, heart and umbilical vein endothelial cells, it is expressed in the highest levels in the brain. Upon brain injury, the extracellular concentrations of nucleotides and cysteinyl leukotrienes (CysLTs), two families of endogenous signaling molecules, increase significantly at the site of damage. In some neurons, GPR17, a membrane receptor for uracil nucleotide and CysLTs, is upregulated as well, infiltrating the lesioned area. GPR17 is thought to play a role in mediating neuronal death, remodeling brain circuitries by microglia and initiating remyelination in damaged neurons. Two named isoforms of GPR17 exist as a result of alternative splicing events.
