Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. The protein encoded by this gene is one of the PKC family members. This kinase has been shown to be involved in many different cellular functions, such as neuron channel activation, apoptosis, cardioprotection from ischemia, heat shock response, as well as insulin exocytosis. Knockout studies in mice suggest that this kinase is important for lipopolysaccharide (LPS)-mediated signaling in activated macrophages and may also play a role in controlling anxiety-like behavior. This gene encodes a protein involved in peripheral nerve myelin upkeep. The encoded protein contains 2 PDZ domains which were named after PSD95 (post synaptic density protein), DlgA (Drosophila disc large tumor suppressor), and ZO1 (a mammalian tight junction protein). Two alternatively spliced transcript variants have been described for this gene which encode different protein isoforms and which are targeted differently in the Schwann cell. Mutations in this gene cause Charcot-Marie-Tooth neuoropathy, type 4F and Dejerine-Sottas neuropathy. [provided by RefSeq, Jul 2008]Seems to be required for maintenance of peripheral nerve myelin sheath. May have a role in axon-glial interactions, possibly by interacting with the cytoplasmic domains of integral membrane proteins such as myelin-associated glycoprotein in the periaxonal regions of the Schwann cell plasma membrane. May have a role in the early phases of myelin deposition.

Clathrin-coated pits and vesicles are assembled for receptor-mediated endocytosis through interaction with clathrin associated protein complexes. Vesicle transport is mediated from the trans-Golgi network by the adapter complex AP-1 and from the plasma membrane by the AP-2 complex. The AP-1 and AP-2 adapter protein complexes consist of clathrin binding adaptin proteins and two smaller subunits known as AP50 and AP17. The Alpha- and Beta-Adaptin chains have a similar two-domain organization with C-terminal domains that vary in both sequence and length. Alpha-Adaptin splice variants A and C display variable relative expression levels and differential distribution in different tissues. AP180 (also designated AP-3 or F1-20) is a synapse-specific clathrin assembly protein. The protein CALM (clathrin assembly protein lymphoid myeloid leukemia) is highly homologous to AP180 and may also be involved in clathrin assembly.

The Annexins are a family of structurally similar proteins. Annexins bind to phospholipids and may be involved in regulation of membrane transport, membrane channel activity, and interaction of the cell membrane with the extracellular matrix. Annexin A4 (ANXA4) belongs to the annexin family of calcium dependent phospholipid binding proteins. Although their functions are still not clearly defined, several members of the annexin family have been implicated in membrane related events along exocytotic and endocytotic pathways. ANXA4 has 45 to 59% identity with other members of its family and shares a similar size and exon intron organization. Isolated from human placenta, ANXA4 encodes a protein that has possible interactions with ATP, and has in vitro anticoagulant activity and also inhibits phospholipase A2 activity. ANXA4 is almost exclusively expressed in epithelial cells.

Mtvr1 is a 176 amino acid protein that exists as two alternatively spliced isoforms. Belonging to the FAM89 family, Mtvr1 is encoded by a gene that maps to human chromosome 11, which comprises approximately 4% of human genomic DNA and is considered a gene and disease association dense chromosome. The chromosome 11 encoded Atm gene is important for regulation of cell cycle arrest and apoptosis following double strand DNA breaks. Atm mutation leads to the disorder known as ataxia-telangiectasia. The blood disorders Sickle cell anemia and thalassemia are caused by HBB gene mutations, while Wilms' tumors, WAGR syndrome and Denys-Drash syndrome are associated with mutations of the WT1 gene. Jervell and Lange-Nielsen syndrome, Jacobsen syndrome, Niemann-Pick disease, hereditary angioedema and Smith-Lemli-Opitz syndrome are also associated with defects in chromosome 11-encoded genes.

Chromosome 1 is the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. When defective, the LMNA gene product can build up in the nucleus and cause characteristic nuclear blebs. The mechanism of rapidly enhanced aging is unclear and is a topic of continuing exploration. The MUTYH gene is located on chromosome 1 and is partially responsible for familial adenomatous polyposis. Stickler syndrome, Parkinson's, Gaucher disease and Usher syndrome are also associated with chromosome 1. A breakpoint has been identified in 1q which disrupts the DISC1 gene and is linked to schizophrenia. Aberrations in chromosome 1 are found in a variety of cancers including head and neck cancer, malignant melanoma and multiple myeloma. The C1orf76 gene product has been provisionally designated C1orf76 pending further characterisation.

FPGT functions to reutilize the L-fucose that is produced uopn glycoprotein and glycolipid turnover.

BAMBI is a membrane spanning glycoprotein that acts as a negative regulator of TGF-beta signaling during development. The BAMBI family of proteins are related to type I TGF-beta receptor family, however, BAMBI is a pseudoreceptor that lacks an intracellular serine/threonine kinase domain. BAMBI transcription regulation is under the influence of beta-catenin, BMP, smad3 and smad4. BAMBI expression can increase in colorectal and hepatocellular carcinomas relative to non-cancerous tissues. BAMBI is expressed at high levels during odontogenesis. BAMBI is coexpressed with Bmp-4 during early Xenopus embryogenesis and can be detected in poorly metastatic human melanoma cell lines.
