Emp is a 396 amino acid ubiquitously expressed adhesion protein. Expressed as 5 alternatively spliced isoforms, Emp contains one CTLH domain and one LisH domain. Emp can form a complex with F-actin, which is involved regulating actin distribution in erythroblasts and macrophages. Considered to assist with cell division and nuclear architecture, Emp is localized with condensed chromatin at prophase, nuclear spindle poles at metaphase and in the contractile ring during telophase and cytokinesis. Although the exact function of Emp is unknown, Emp is suggested to be involvement in erythroblast-macrophage cell attachment, terminal maturation and enucleation of erythroid cells, and inhibiting apoptosis of erythroblasts.

Serotonin (5-hydroxytryptamine, 5-HT), a neurotransmitter, elicits a wide array of physiological effects by binding to several receptor subtypes, including the 5-HT2 family of seven-transmembrane-spanning, G-protein-coupled receptors, which activate phospholipase C and D signaling pathways. This gene encodes the 2C subtype of serotonin receptor and its mRNA is subject to multiple RNA editing events, where genomically encoded adenosine residues are converted to inosines. RNA editing is predicted to alter amino acids within the second intracellular loop of the 5-HT2C receptor and generate receptor isoforms that differ in their ability to interact with G proteins and the activation of phospholipase C and D signaling cascades, thus modulating serotonergic neurotransmission in the central nervous system. Studies in humans have reported abnormalities in patterns of 5-HT2C editing in depressed suicide victims. [provided by RefSeq, Jul 2008].

The homeobox is DNA sequence that is approximately 180 base pairs long that is involved in all steps of the developmental regulation of animals, fungi and plants, from embryogenesis to cell differentiation. The homeobox encodes a protein domain of about 60 amino acids, called the homeodomain, that can bind to DNA. The gastrulation brain homeobox 1 (GBX1) gene, along with another homeobox gene (EN2), maps to chromosome 7q36, and is expressed in septal and pallidal areas and in the basal forebrain cholinergic system, an area implicated in Alzheimer’s disease. The GBX1 gene encodes the GBX1 protein, which localizes to the nucleus and contains 1 homeobox DNA-binding domain.

This gene encodes a ubiquitous nuclear protein that stimulates both cell proliferation and differentiation. It is a member of the MADS (MCM1, Agamous, Deficiens, and SRF) box superfamily of transcription factors. This protein binds to the serum response element (SRE) in the promoter region of target genes. This protein regulates the activity of many immediate-early genes, for example c-fos, and thereby participates in cell cycle regulation, apoptosis, cell growth, and cell differentiation. This gene is the downstream target of many pathways; for example, the mitogen-activated protein kinase pathway (MAPK) that acts through the ternary complex factors (TCFs). ANKRD54 is a 300 amino acid protein that contains four ankyrin repeats and is expressed as two alternatively spliced isoforms. The gene encoding human ANKRD54 maps to chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, neurofibromatosis type 2, autism and schizophrenia. Additionally, translocations between chromosomes 9 and 22 may lead to the formation of the Philadelphia chromosome and the subsequent production of the novel fusion protein BCR-Abl, a potent cell proliferation activator found in several types of leukemias.

Cell adhesion molecules (CAMs) influence cell growth, differentiation, embryogenesis, immune response and cancer metastasis by networking information from the extracellular matrix to the cell. The four major families of cell adhesion molecules are immunoglobulin (Ig) superfamily (calcium-independent transmembrane glycoproteins), integrins (transmembrane non-covalently linked heterodimers of Alpha and Beta subunits), calcium-dependent cadherins and divalent cation-dependent selectins. Regulation of neuronal synaptic adhesion by CAMs has proven important for learning and memory. Proper embryonic morphogenic development is also heavily dependent on the regulation of cell adhesion molecules. Neurotrimin (hNT) is a neural cell adhesion molecule localizing to the cell membrane, where it acts as a lipid-anchor. Neurotrimin belongs to the IgLON family of proteins, a member of the larger immunoglobulin superfamily.

ATRNL1 is a 1,379 amino acid single-pass type I membrane protein that may play a role in melanocortin signaling pathways that regulate energy homeostasis. The ATRNL1 protein contains a C-type lectin domain, a CUB domain, two EGF-like domains, six Kelch repeats, two laminin EGF-like domains and five PSI domains. ATRNL1 interacts with MC4-R in several regions known to be important in the regulation of energy homeostasis by melanocortins, such as the paraventricular nucleus of hypothalamus and the dorsal motor nucleus of the vagus. The ATRNL1 gene is conserved in dog, cow, mouse, rat, chicken, zebrafish and C. elegans, exists as two alternatively spliced isoforms and maps to human chromosome 10q25.3. Strong evidence of linkage to late-onset Alzheimer disease (LOAD) is linked to chromosome 10, which implicates a wide region and at least one disease-susceptibility locus.

Gamma-aminobutyric acid type A (GABAA) receptors mediate inhibitory neurotransmission in the mammalian central nervous system. The receptor exists as a pentameric ion channel composed by heteromeric combinations of alpha, beta, gamma, delta, epsilon, theta, or pi subunits. Only specific subunit combinations produce viable receptors, while others never translocate to the cell surface from the ER where they are synthesised, and subsequently degraded. The theta subunit forms a receptor in combination with alpha3 subunits in monoaminergic cell groups. These receptors, found especially in the septum, preoptic areas, hypothalamic nuclei, amygdala and thalamus, likely have unique pharmacological properties linked to their expression in this particular cell type and not cholinergic cell groups, and may play a role in opiate withdrawal symptoms.

E3 ubiquitin-protein ligase component of the LUBAC complex which conjugates linear ('Met-1'-linked) polyubiquitin chains to substrates and plays a key role in NF-kappa-B activation and regulation of inflammation. LUBAC conjugates linear polyubiquitin to IKBKG and RIPK1 and is involved in activation of the canonical NF-kappa-B and the JNK signaling pathways. Linear ubiquitination mediated by the LUBAC complex interferes with TNF-induced cell death and thereby prevents inflammation. LUBAC is proposed to be recruited to the TNF-R1 signaling complex (TNF-RSC) following polyubiquitination of TNF-RSC components by BIRC2 and/or BIRC3 and to conjugate linear polyubiquitin to IKBKG and possibly other components contributing to the stability of the complex. Together with FAM105B/otulin, the LUBAC complex regulates the canonical Wnt signaling during angiogenesis. Binds polyubiquitin of different linkage types.
