This antibody reacts with the extracellular domain of EGFR and blocks the EGF/TGF alpha induced activation. It also blocks tumor growth in vivo. It is excellent for purification of EGFR. EGFR is type I receptor tyrosine kinase with sequence homology to erbB-1, -2, -3 -4 or HER-1, -2, -3 -4. It binds to Epidermal Growth Factor (EGF), Transforming Growth Factor-a (TGF-a), Heparin-binding EGF (HB-EGF), amphiregulin, βcellulin and epiregulin.

This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.

Recognizes a protein of 50-65 kDa, identified as CD16 (Workshop IV; Code N39 ) (also known low affinity Fc receptor III for IgG (FcRIII) or Leu 11). CD16 exists as a polypepetide-anchored from (FCRIIIA or CD16A) on human natural killer (NK) cells and monocytes/ macrophages and as a glycosylphosphatidylinositol (GPI)-anchored form (FcRIIIB or CD16B) on neutrophils. CD16B is polymorphic and the two alleles are termed NA1 and NA2.3 CD16 plays a role in signal transduction, NK cell activation and antibody-dependent cellular cytotoxicity. This MAb has been showed to inhibit the binding of immune complex to NK cells, inhibit cytotoxicity of NK cells, and induce calcium fluxes in NK cells and neutrophils.

This antibody recognizes a protein of 54 kDa, which is identified as AMACR, also known as p504S. It is an enzyme that is involved in bile acid biosynthesis and β-oxidation of branched-chain fatty acids. AMACR is essential in lipid metabolism. It is expressed in cells of premalignant high-grade prostatic intraepithelial neoplasia (HGPIN) and prostate adenocarcinoma. The majority of the carcinoma cells show a distinct granular cytoplasmic staining reaction. AMACR is present at low or undetectable levels in glandular epithelial cells of normal prostate and benign prostatic hyperplasia. A spotty granular cytoplasmic staining is seen in a few cells of the benign glands. AMACR is expressed in normal liver (hepatocytes), kidney (tubular epithelial cells) and gall bladder (epithelial cells). Expression has also been found in lung (bronchial epithelial cells) and colon (colonic surface epithelium). AMACR expression can also be found in hepatocellular carcinoma and kidney carcinoma. Past studies have also shown that AMACR is expressed in various colon carcinomas (well, moderately and poorly differentiated) and overexpressed in prostate carcinoma.

This MAb reacts specifically with heat shock protein HSP27 in human and monkey tissues and cell lines such as MCF-7. HSP27, also referred to as the Estrogen-Regulated 24K protein and HSP28, is one of several small heat shock proteins produced by all organisms studied. HSP27 synthesis is induced by elevated temperature, as well as by estrogen in hormone responsive cells. Interestingly, human HSP27 also shares greater than 50% homology with low molecular weight Drosophila HSPs and mammalian α-crystalline lens protein. Because of the estrogen responsive nature of HSP27, this protein has been studied extensively in human estrogen responsive tissues such as cervix, endometrium and breast tissue. Therefore HSP27 may be useful in classifying various hormone sensitive tumors.

This antibody recognizes a 24-27 kDa estrogen-regulated protein, identified as heat shock protein 27 (hsp27). Hsp27 was recently found to be identical to the estrogen-induced p29 and 24K protein. About 50% of breast carcinomas are positive for hsp27 especially those that are also positive for estrogen and/or progesterone receptor. HSP27 has also been implicated in drug resistance in cancer cells.

Recognizes a protein of 94 kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. Grp94 has been briefly studied as a prognostic factor in breast cancer.

CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

The EBV-induced early antigens (Ea) are among several antigen complexes that have been identified in EBV-infected cells. The Ea complex is composed of diffuse (Ea-D) and restricted (Ea-R) components. The activity of Ea-D is suppressed during latent infection. BMRF1, the gene that encodes for Ea-D, is closely associated with the gene encoding for EBV DNA polymerase, and Ea-D is essential for the activity of this polymerase. Ea-D forms a complex with EBV DNase and, together, they may play a role in viral replication.
