In Western blotting, this antibody recognizes proteins in MW range of 265-400 kDa, identified as different glycoforms of EMA. EMA may provide a protective layer on epithelial cells against bacterial and enzyme attack. In immunohistochemical assays, it superbly stains routine formalin/paraffin carcinomas. Antibody to EMA is useful as a pan-epithelial marker for detecting early metastatic loci of carcinoma in bone marrow or liver.

Granule proteins are released by physiologic and pharmacologic stimuli and play important roles in both normal and pathological host immune responses. Eosinophil major basic protein and eosinophil peroxidase (EPX) are granule proteins specific to the eosinophil. AHE-1 recognizes human EPX, a granule protein specific to eosinophils. It does not cross-react with eosinophil major basic protein, elastase, cathepsin G, esterase N, thrombin, plasmin, kallikrein, lactoferrin, or transferrin. This MAb stains eosinophils only and does not stain other peripheral blood cells, including platelets, neutrophils, monocytes, lymphocytes or red blood cells. Human EPX gene product can form a tetramer of two light chains and two heavy chains. Other peroxidase family members include myeloperoxidase (MPO), lactoperoxidase (LPO), and thyroid peroxidase (TPO).

This MAb is specific to ER alpha and shows minimal cross-reaction with other members of the family. ER is an important regulator of growth and differentiation in the mammary gland. Presence of ER in breast tumors indicates an increased likelihood of response to anti-estrogen (e.g. tamoxifen) therapy. This MAb is excellent for staining of formalin-fixed, paraffin-embedded breast carcinomas.

Recognizes an oncofetal antigen of 220 kDa, identified as a tumor-associated glycoprotein (TAG-72) with properties of a mucin. This MAb defines the mucin-carried sialylated-Tn epitope. TAG-72 is usually expressed by adenocarcinomas, but is negative in mesotheliomas. Studies have reported that this antibody has 80% sensitivity and 93% specificity for pulmonary adenocarcinoma. Therefore, TAG-72 is a useful marker to distinguish between mesothelioma and adenocarcinoma. However, false positive reactions can occur so results must be interpreted with the utmost caution. This antibody may be useful in the differentiation of non-small cell carcinomas from small cell carcinomas of the lung. The combined use of anti-TAG-72 and anti-GCDFP-15 is valuable in the diagnosis of apocrine carcinoma.

Recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. This MAb selectively interferes with lymphocyte binding to lymph node, mucosal and synovial endothelium. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. CD44 immunostaining is commonly used for the discrimination of urothelial transitional cell carcinoma in-situ from non-neoplastic changes in the urothelium.

Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

Recognizes a sialoglycoprotein of 27-32 kDa, identified as CD99, or MIC2 gene product, or E2 antigen. MIC2 gene is located in the pseudo-autosomal region of the human X and Y chromosome. MIC2 gene encodes two distinct proteins, which are produced by alternative splicing of the CD99 gene transcript and are identified as bands of 30 and 32 kDa (p30/32).Although its function is not fully understood, CD99 is implicated in various cellular processes including homotypic aggregation of T cells, upregulation of T cell receptor and MHS molecules, apoptosis of immature thymocytes and leukocyte diapedesis.CD99 is expressed on the cell membrane of some lymphocytes, cortical thymocytes, and granulosa cells of the ovary. Most pancreatic islet cells, Sertoli cells of the testis, and some endothelial cells express this antigen. Mature granulocytes express very little or no CD99. MIC2 is strongly expressed on Ewing's sarcoma cells and primitive peripheral neuroectodermal tumors. This MAb shows a very similar reactivity to other CD99 MAbs (e.g. O13, 12E7, or HBA-71) and is excellent for immunohistochemical staining of formalin-fixed, paraffin-embedded tissues.

Recognizes the -chain of CD3, which consists of five different polypeptide chains (designated as γ, δ, ε, ζ, and η) with MW ranging from 16-28 kDa. The CD3 complex is closely associated at the lymphocyte cell surface with the T cell antigen receptor (TCR). Reportedly, CD3 complex is involved in signal transduction to the T cell interior following antigen recognition. The CD3 antigen is first detectable in early thymocytes and probably represents one of the earliest signs of commitment to the T cell lineage. In cortical thymocytes, CD3 is predominantly intra-cytoplasmic. However, in medullary thymocytes, it appears on the T cell surface. CD3 antigen is a highly specific marker for T cells, and is present in majority of T cell neoplasms.

CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD45R functions as a phosphor-tyrosine phosphatase. This MAb reacts with all isoforms of CD45R expressed by all hematopoietic cells, except erythrocytes, having a higher level of expression on lymphocytes than on granulocytes (Workshop IV). Epitope of this MAb resides in the sialic acid moiety of T200 molecule. It enhances the proliferative response of B cells in an anti-immunoglobulin-induced B cell proliferation. Antibody to CD45 is useful in differential diagnosis of lymphoid tumors from non-hematopoietic undifferentiated neoplasms.

The MUC6 gastric mucin is a secreted glycoprotein that plays an essential role in epithelial cyto-protection from acid, proteases, pathogenic microorganisms, and mechanical trauma in the gastrointestinal tract. Mucin 6 expression is highest in the stomach and gall bladder, with lower expression in the terminal ileum and right colon. In gastric cancer, Mucin 6 has an altered expression. In normal stomach, Mucin 6 is associated with Lewis type 2; Mucin 6 is also expressed in gastric metaplasia, duodenum and pancreas. Mucin 6 is a secretory mucin, located in the deeper mucosal folds of human gall bladder, and its expression is altered with increasing degrees of inflammation.
