AHNAK is a 5,890 amino acid protein encoded by the human gene AHNAK. The intronless AHNAK gene is located on human chromosome 11q12 and has three main structural regions: the 251 amino acid N-terminus, a large central region of 4390 amino acids with multiple repeated units of about 128 amino acids in length, and the 1002 amino acid C-terminus. The central region seems to have antiparallel beta-strands connected by intervening loops. Several putative regulatory elements are clustered within the C-terminal region, including nuclear export localization signals, a leucine zipper, and potential phosphorylation sites for Akt1 and PKC. AHNAK is believed to be an important signalling molecule involved in a wide range of physiological activities and may be required for neuronal cell differentiation.. AHNAK also appears to influence b-adrenergic regulation of cardiac L-type Ca2+ channel function.

GPRC6A (G protein-coupled receptor family C group 6 member A) is a 928 amino acid protein encoded by the mouse Gprc6a gene. GPRC6A is a member of the G protein-coupled receptor 3 family. This receptor is activated by both amino acids and extracellular concentration of calcium ions. The activity of GPRC6A is mediated by a G protein that activates a phosphatidylinositol-calcium second messenger system. GPRC6A senses changes in the extracellular concentration of calcium ions, suggesting that it may mediate extracellular calcium-sensing responses in osteoblasts. Osteocalin stimulates the activity of GPRC6A in the presence of calcium. GPRC6A also acts as a receptor for amino acids, with a preference for basic amino acids such as L-Lys, L-Arg and L-ornithine. Its affinity for amino acids suggests that it may act as a regulatory component of the urea cycle. GPRC6A is expressed at high levels in liver, lung, spleen and heart, and at lower levels in kidney, skeletal muscle and brain.

The transmembrane semaphorins are characterized by an additional 80 amino acid transmembrane domain and an 80-110 amino acid cytoplasmic domain. Secreted and cell-bound semaphorins chemically attract and repel the growth of neural axons, guiding the development of intricate networks of neural tissue. SEMA3E is a secreted semaphorin with 775 amino acids. Mutations in the SEMA3E gene are associated with CHARGE syndrome, a disorder characterized by cranial nerve dysfunction, coloboma of the eye, choanal atresia, inner and external ear abnormalities, cardiac anomalies, genitourinary abnormalities, and growth retardation.

The extracellular domain of SEMA6C induces growth cone collapse of dorsal root ganglion and plays a role in generation or stability of entorhino-hippocampal synapses.
