CD99 is a type I transmembrane glycoprotein and the founding member of the CD99 family of molecules. The extracellular domain of CD99 contains no identifiable motifs, its cytoplasmic region, although short, does have signal transduction capability. Cells known to express CD99 include fibroblasts, neutrophils, T cells, double positive thymocytes, CD34+ stem cells, monocytes and endothelial cells. Two types of CD99 isoforms have been classified. Native human CD99 is referred to as the long, or type I isoform. The best studied type II isoform shows an Asp-Gly substitution for the C terminal 27 amino acids. The type I and II isoforms have distinctive signal transduction pathways (FAKsrc for type I PI3K plus srcERK1/2 for type II), and mediate clearly different biological outcomes. Homophilic interaction between CD99 on the neutrophil and CD99 on the endothelial cell regulates the transendothelial migration of neutrophils during inflammation. Human CD99 has 48% aa sequence identity to mouse CD99.

Activin receptor type-1, also known as Activin receptor type I, Activin receptor-like kinase 2, Serine/threonine-protein kinase receptor R1, TGF-B superfamily receptor type I, ACVRLK2 and ACVR1, is a single-pass type I membrane protein. ACVR1 is expressed in normal parenchymal cells, endothelial cells, fibroblasts and tumor-derived epithelial cells. ACVR1 belongs to the protein kinase superfamily. Activins signal through a heteromeric complex of receptor serine kinases which include at least two type I (I and IB) and two type II (II and IIB) receptors. These receptors are all transmembrane proteins, composed of a ligand-binding extracellular domain with cysteine-rich region, a transmembrane domain, and a cytoplasmic domain with predicted serine/threonine specificity. Type I receptors are essential for signaling; and type II receptors are required for binding ligands and for expression of type I receptors. Type I and II receptors form a stable complex after ligand binding, resulting in phosphorylation of type I receptors by type II receptors. ACVR1 signals a particular transcriptional response in concert with activin type II receptors.

Vaspin (Visceral Adipose-Specific SERPIN) is a newly described adipokine. Vaspin has three beta -sheets, nine alpha-helices, and one central loop; the structure is part of the set of distinctive features that are descriptive of Serpin family members. Vaspin is also a unique insulin sensitizing adipocytokine in obesity. A recent publication indicates that Vaspin mRNA expression in visceral fat is positively correlated with BMI and percent of body fat. and could be associated with parameters of obesity, insulin resistance, and glucose metabolism. These findings suggest a potential clinical use for Vaspin in ameliorating certain aberrations seen in the obesity metabolic syndrome.

The 504 amino acid human SIRP alpha contains two Ig-like C1-type domains and one Ig-like V-type domain. SIRP alpha can express in various tissues, mainly on brain and myeloid cells, including macrophages, neutrophils, dendritic and Langerhans cells. It also can detect in neurons, smooth muscle and endothelial cells. SIRPA is an immunoglobulin-like cell surface receptor for CD47. MHC Class I Polypeptide-Related Sequence A (MICA) is a transmembrane glycoprotein that functions as a ligand for human NKG2D. Unlike classical MHC class I molecules, MICA does not form a heterodimer with beta-2-microglobulin. MICA shares 85% amino acid identity with a closely related protein, MICB. MICA acts as a stress-induced self-antigen that is recognized by NK cells, NKT cells, and most of the subtypes of T cells. As a Ligand for the KLRK1/NKG2D receptor, MICA binds to KLRK1 leads to cell lysis. MICA functions as an antigen for gamma delta T cells and is frequently expressed in epithelial tumors. MICA antigens are able to elicit the synthesis of alloantibodies in transplant recipients. Studies have shown that anti-MICA antibodies are associated with acute renal allograft rejection and failure. MICA recognition is involved in tumor surveillance, viral infections, and autoimmune diseases.
