Rab17 belongs to the Rab family of small Ras-like GTPases. It is specifically expressed in epithelial cells and is upregulated during cell polarization. Immunofluorescence staining studies indicate that Rab17 is associated with the perinuclear recycling endosome in nonpolarized epithelial cells and with the apical recycling endosome in polarized epithelial cells. The function of Rab17 remains unclear. Reports of Rab17 colocalization with internalized IgA in the apical endosome suggest that it may regulate receptor-mediated transcytosis. Rab17 has also been shown to regulate melanocytic filopodia formation and melanosome trafficking. siRNA knockdown of Rab17 in melanoma cells induces melanosome accumulation in the cell periphery.

SART1 encodes two proteins, the SART1(800) protein expressed in the nucleus of the majority of proliferating cells, and the SART1(259) protein expressed in the cytosol of epithelial cancers. The SART1(259) protein is translated by the mechanism of -1 frameshifting during posttranscriptional regulation; its full length sequence is not published yet. The two encoded proteins are thought to be involved in the regulation of proliferation. Both proteins have tumor rejection antigens. The SART1(259) protein possesses tumor epitopes capable of inducing HLA-A2402-restricted cytotoxic T lymphocytes in cancer patients. This SART1(259) antigen may be useful in specific immunotherapy for cancer patients and may serve as a paradigmatic tool for the diagnosis and treatment of patients with atopy. The SART1(259) protein is found to be essential for the recruitment of the tri snRNP to the pre spliceosome in the spliceosome assembly pathway.

The human Mel-CAM gene maps to chromosome 11q23 and encodes a trans-membrane glycoprotein, also designated MCAM, MUC 18 or CD146, that belongs to the immunoglobulin superfamily and functions as a Ca2 -independent cell adhesion molecule. Mel-CAM expression is restricted to advanced primary and metastatic melanomas and to cell lines of the neuroectodermal lineage, but not normal melanocytes. Mel-CAM is found on 80% of advanced primary human melanomas and correlates well with development of metastatic disease.

C3orf32 (chromosome 3 open reading frame 32), also known as fls485, is a 353 amino acid protein encoded by a gene that maps to human chromosome 3p26.1. Chromosome 3 is made up of approximately 214 million bases encoding over 1,100 genes. Notably, there is a chemokine receptor gene cluster and a variety of human cancer related loci on chromosome 3. Particular regions of the chromosome 3 short arm are deleted in many types of cancer cells. Key tumor suppressing genes on chromosome 3 encode apoptosis mediator RASSF1, cell migration regulator HYAL1 and angiogenesis suppressor SEMA3B. PED/PEA 15 (Phosphoprotein Enriched in Diabetes/Phosphoprotein Enriched in Astrocytes 15 kDa) is a widely expressed 15 kDa protein comprised of an N terminal region containing a canonical Death Effector Domain (DED) sequence and a nuclear export signal, and a C terminal region containing two serine phosphorylation sites. PED/PEA 15 has been implicated in the regulation of multiple cellular processes including apoptosis, integrin activation, and insulin sensitive glucose transport in insulin responsive cells. Phosphorylation of both serine 104 (a Protein Kinase C site) and serine 116 (a substrate of CaMKII and Akt) is required for PED/PEA 15 function.

Pax genes contain paired domains with strong homology to genes in Drosophila, which are involved in programming early development. Lesions in the Pax-6 gene account for most cases of aniridia, a congenital malformation of the eye, chiefly characterized by iris hypoplasia, which can cause blindness. Pax-6 is involved in other anterior segment malformations besides aniridia, such as Peters anomaly, a major error in the embryonic development of the eye with corneal clouding with variable iridolenticulocorneal adhesions. The Pax-6 gene encodes a transcriptional regulator that recognizes target genes through its paired-type DNA-binding domain. The paired domain is composed of two distinct DNA-binding subdomains, the amino-terminal subdomain and the carboxy-terminal subdomain, which bind respective consensus DNA sequences. The human Pax-6 gene produces two alternatively spliced isoforms that have the distinct structure of the paired domain.
