The protein encoded by this gene is one of several proteins that interact with NEDD4, a member of a family of ubiquitin-protein ligases. These proteins have PY motifs in common that bind to the WW domains of NEDD4. NEDD4 is developmentally regulated, and is highly expressed in embryonic tissues. Mutations in this gene (i.e., intronic insertions of >100 copies of pentanucleotide repeats including a (TGGAA)n sequence) are associated with spinocerebellar ataxia type 31. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2010].

Recognizes a protein of 150 kDa, which is identified as the high molecular weight variant of Caldesmon. Two closely related variants of human caldesmon have been identified which are different in their electrophoretic mobility and cellular distribution. The h-caldesmon variant (120-150 kDa) is predominantly expressed in smooth muscle whereas l-caldesmon (70-80 kDa) is found in non- muscle tissue and cells. Neither of the two variants has been detected in skeletal muscle. This MAb recognizes only the 150 kDa variant (h-caldesmon) in Western blots of human aortic media extracts and is unreactive with fibroblast extracts from cultivated human foreskin. Caldesmon is a developmentally regulated protein involved in smooth muscle and non-muscle contraction.

GDE1 is a 331 amino acid multi-pass membrane protein that localizes to both the membrane and the cytoplasm and contains one GDPD domain. Expressed in a wide variety of tissues, GDE1 uses magnesium as a cofactor to catalyse the conversion of 1-(sn-glycero-3-phospho)-1D-myo-inositol to myo-inositol and sn-glycerol 3-phosphate, an event that is modulated by G protein Signalling pathways and provides a link between phosphoinositide metabolism and G protein signal transduction. The gene encoding GDE1 maps to human chromosome 16, which encodes over 900 genes and comprises nearly 3% of the human genome. The GAN gene is located on chromosome 16 and, with mutation, may lead to giant axonal neuropathy, a nervous system disorder characterised by increasing malfunction with growth. The rare disorder Rubinstein-Taybi syndrome is also associated with chromosome 16, as is Crohn's disease, which is a gastrointestinal inflammatory condition.

Beta-defensins (also designated BD, and hBD in human) are small cationic peptides with broad-spectrum antimicrobial activity. Produced in mucosal epithelia and neutrophils of several species, Beta-defensins are developmentally regulated. Human b-defensin 2 is locally regulated by inflammation and is the first member of the b-defensin family that is locally inducible by inflammation. The murine homolog of human b-defensin 2, which is called b-defensin 3, is present in the respiratory system and in low levels in the epithelial cells of the intestine and lung. The unique murine b-defensin 2 (Defb2) is not expressed in airways of untreated mice, but is upregulated in the airways by lipopolysaccharide and may contribute to host defense at the mucosal surface of the airways
