MultimericCD40L™ is a high activity construct in which two trimeric CD40 ligands are artificially linked via the collagen domain of ACRP30. This construct very effectively simulates the natural membrane-assisted aggregation of CD40L in vivo. It provides a simple and equally potent alternative to CD40L+enhancer combinations. MultimericCD40L™ has shown to suppress alum-induced IL-1beta release and caspase-1 activation in a dose-, CD40- and time dependent manner, without affecting BMDM (Bone marrow-derived macrophages) viability. It also effectively suppressed the inflammasome function triggered by NLRP3 activators. The secretion of caspase-1 independent inflammatory mediators has been shown to be unaltered or even enhanced.

Four human beta -Defensins have been identified to date: BD-1, BD-2, BD-3 and BD-4. beta -defensins contain a six-cysteine motif, they forms three intra-molecular disulfide bonds. beta -defensins are also chemoattractant towards immature dendritic cells and memory T cells. The beta -defensin proteins are expressed as the C-terminal portion of precursors; they are released by proteolytic cleavage of a signal sequence.

Insulin-like growth factors (IGFs) comprise a family of endocrine, paracrine and autocrine polypeptides consisting of the ligands IGF1 and IGF2, two receptors (IGF1R, IGF2R), at least 6 IGF-binding proteins (IGFBPs) and IGFBP proteases. Among the binding proteins, IGFBP6 is unique because of its N-terminal disulphide linkages and its marked binding preference for IGF2. It is a potent inhibitor of the interaction between IGF2 and its receptor IGF1R, thus preventing major functions of IGF2, such as induction of proliferation, differentiation, cell adhesion, or colony formation. In particular, IGFBP-6 inhibited the growth of neuroblastoma and rhabdomyosarcoma xenografts. GFBP-6 is expressed in many tissues, including lung, liver, gut and the central nervous system.

Human Chemokine (C-X-C motif) Ligand 7 (CXCL7), also known as neutrophil activating peptide 2 (NAP-2), is a member of the CXC chemokines containing an ELR domain (Glu-Leu-Arg tripeptide motif). Similar to other ELR domain containing CXC chemokines, such as IL-8 and the GRO proteins, CXCL7 binds CXCR2, chemoattracts and activates neutrophils. CXCL7, Connective Tissue Activating Protein III (CTAPIII) and beta thrombogulin ( beta TG), are proteolytically processed carboxylterminal fragments of platelet basic protein (PBP) which is found in the alphagranules of human platelets. Although CTAPIII, beta TG, and PBP represent amino-terminal extended variants of NAP2 and possess the same CXC chemokine domains, these proteins do not exhibit CXCL7/NAP2 activity. CXCL7 induces cell migration through the G-protein-linked receptor CXCR-2.

Signal Transducer and Activator of Transcription 5b (STAT5B) is a member of the STAT family of transcription factors. They are responsible for an array of cellular activities including regulating growth, survival, differentiation, motility, and the immune response. STAT5B mediates the signal transduction triggered by various cell ligands, such as IL2, IL4, CSF1, and different growth hormones. It has been shown to be involved in diverse biological processes, such as TCR signalling, apoptosis, adult mammary gland development, and sexual dimorphism of liver gene expression. Signal transduction and activator of transcription 5 (STAT5) is a member of the Jak/STAT signal transduction pathway and is activated by a variety of cytokines (IL22, IL6). STAT5 has two isoforms (A and B) that share 93% amino acid identity and bind the DNA consensus site TTCN3GAA. STAT5 mediates cytokine signalling by acting as a signal transducer in the cytoplasm and, upon phosphorylation, translocates to the nucleus and activates transcription of specific genes. STAT5 is involved in a wide array of biological processes ranging from regulating apoptosis to adult mammary gland proliferation, differentiation and survival.
