Interleukin-20 Receptor Subunit alpha (IL20RA) is a single-pass type I membrane protein that is a member of the type II cytokine receptor family. IL20RA is synthetized a 553 amino acid glycoprotein precursor containing a 29 amino acid signal peptide, a 221 amino acid extracellular domain with two fibronectin type-III domains, a 24 amino acid transmembrane region, and a 279 amino acid intracellular domain. IL20RA is widely expressed with highest levels found in skin and testis and high levels in brain. IL20RA forms a heterodimer with IL20RB, and the complex serves as a receptor for IL19, IL20 and IL24. IL20RA also forms a heterodimer with the unique and specific receptor IL10RB and functions as the receptor for IL26.

Insulin-Degrading Enzyme (IDE) is a secreted enzyme that belongs to the peptidase M16 family. IDE is a large zinc-binding protease and cleaves multiple short polypeptides that vary considerably in sequence. IDE plays a role in the cellular breakdown of insulin, IAPP, glucagon, bradykinin, kallidin, and other peptides, and thereby plays a role in intercellular peptide signaling. IDE degrades amyloid formed by APP and IAPP. IDE may participate in the degradation and clearance of naturally secreted amyloid beta -protein by neurons and microglia. IDE, which migrates at 110 kDa during gel electrophoresis under denaturing conditions, has since been shown to have additional substrates, including the signaling peptides glucagon, TGF alpha and beta -endorphin.

B7-H4 is a B7 family member that negatively regulates T cell immunity by inhibiting of T cell proliferation, cytokine production and cell cycle progression. In vivo, blockade of endogenous B7-H4 by specific monoclonal antibody promotes T cell responses. B7-H4 ia an important negative regulator of innate immunity through growth inhibition of neutrophils. B7-H4 is expressed on some tumour cancer cells. The role of B7-H4 in tumour progression may be to transform precancerous cells and then protect them from immunosurveillance.

BAFF is mainly produced by innate immune cells such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells. T cells, activated B cells, some malignant B cells and also non-lymphoid cells like astrocytes, synoviocytes and epithelial cells can also produce BAFF. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation. Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of BAFF leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases such as Sjoegren syndrome, Rheumatoid arthritis (RA), Multiple sclerosis (MS) and Systemic Lupus Erythematosus (SLE). BAFF has also increased levels in some lymphoid cancers.
