It is a single-pass type I membrane protein and contains 4 TNFR-Cys repeats. The protein is a member of the tumor necrosis factor (TNF) family of receptors. It is expressed on the surface of most cell types, including cells of epithelial and myeloid lineages, but not on T and B lymphocytes. The protein is the receptor for the heterotrimeric lymphotoxin containing LTA and LTB, and for TNFS14/LIGHT. It promotes apoptosis via TRAF3 and TRAF5 and may play a role in the development of lymphoid organs. The encoded protein and its ligand play a role in the development and organization of lymphoid tissue and transformed cells. Activation of the encoded protein can trigger apoptosis. Not only does the TNFRSF3 help trigger apoptosis, it can lead to the release of the cytokine interleukin 8. Overexpression of TNFRSF3 in Human Cells cells increases IL-8 promoter activity and leads to IL-8 release. TNFRSF3 is also essential for development and organization of the secondary lymphoid organs and chemokine release.

Tyrosine-protein kinase transmembrane receptor ROR1 is also known as Neurotrophic tyrosine kinase, receptor-related 1 (NTRKR1), which belongs to the protein kinase superfamily or tyr protein kinase family or ROR subfamily. ROR1 contains 1 FZ (frizzled) domain, 1 Ig-like C2-type (immunoglobulin-like) domain, 1 kringle domain, 1 protein kinase domain. ROR1 is expressed at high levels during early embryonic development. The expression levels drop strongly around day 16 and there are only very low levels in adult tissues. Isoform Short is strongly expressed in fetal and adult CNS and in a variety of human cancers, including those originating from CNS or PNS neuroectoderm. ROR1 could interact with casein kinase 1 epsilon (CK1ε) to activate phosphoinositide 3-kinase-mediated AKT phosphorylation and cAMP-response-element-binding protein (CREB), which was associated with enhanced tumor-cell growth.

4-1BB is also known as CD137, tumor necrosis factor receptor superfamily member 9 (TNFRSF9), induced by lymphocyte activation (ILA), is a co-stimulatory molecule of the tumor necrosis factor (TNF) receptor superfamily. CD137 can be expressed by activated T cells, but to a larger extent on CD8 than on CD4 T cells. In addition, CD137 expression is found on dendritic cells, follicular dendritic cells, natural killer cells, granulocytes and cells of blood vessel walls at sites of inflammation. The best characterized activity of CD137 is its costimulatory activity for activated T cells. Crosslinking of CD137 enhances T cell proliferation, IL-2 secretion survival and cytolytic activity. Further, it can enhance immune activity to eliminate tumors in mice. CD137 can enhance activation-induced T cell apoptosis when triggered by engagement of the TCR/CD3 complex. In addition, 4-1BB/4-1BBL co-stimulatory pathway has been shown to augment secondary CTL responses to several viruses, and meanwhile augment anti-tumor immunity. 4-1BB thus is a promising candidate for immunotherapy of human cancer. CD137 has been shown to interact with TRAF2.

Leukocyte-Associated Immunoglobulin-Like Receptor 2 (LAIR2) is a secreted, 131 amino acid protein that contains one Ig-like C2 type domain, making it a member of the Ig superfamily. When compared to LAIR-1, its transmembrane counterpart, it shares 83% amino acid identity across the signal sequence and extracellular domains; although one is secreted and one is membrane-bound, the two LAIR proteins are thought to have arisen from a common gene ancestor and appear to share similar adhesion profiles. This suggests that LAIR-2 may compete with LAIR-1 for ligand binding. A 114 amino acid alternate splice form of LAIR-2 is truncated at the C terminus, but retains the entire Ig domain. The expression profile of these splice forms, and the presence of orthologs in other species, have not been reported.

CD86 (B7-2) is a 60-100 kDa variably glycosylated protein in the B7 family. B7 family members are transmembrane cell surface molecules that play important roles in immune activation and the maintenance of immune tolerance. B7-2 is highly expressed on activated antigen presenting cells (APC), e.g. B cells, dendritic cells and monocytes as well as on vascular endothelial cells. B7-2 and the closely related CD80 (B7-1) exhibit overlapping but distinct functional properties. Their binding to CD28, which is constitutively expressed on T cells, enhances T cell receptor signalling and also provides TCR-independent costimulation. B7-1 and B7-2 additionally bind the CD28-related protein CTLA-4, which is up-regulated and recruited to the immunological synapse (IS) at the onset of T cell activation. CTLA-4 ligation inhibits the T cell response and supports regulatory T cell function. B7-2 is expressed earlier than B7-1 following APC activation and both proteins bind with higher affinity to CTLA-4 than to CD28. B7-2 promotes the stabilisation of CD28 in the IS, while B7-1 is primarily responsible for promoting CTLA-4 recruitment and accumulation in the IS. The relative participation of B7-1 and B7-2 in T cell costimulation can also alter the Th1/Th2 bias of the immune response. Both B7-1 and B7-2 serve as cellular receptors for B species adenoviruses.

IL6ST /gp130 /CD130 can be found in tissues such as brain, heart, thymus, spleen, kidney, lung and liver and found in all the cell lines tested except BaF-B03.The expression of IL-6ST /gp130 is not restricted to IL6-responsive cells. The receptor systems for IL6, LIF, OSM, CNTF, IL11, CTF1 and BSF3 can utilize gp130 for initiating signal transmission. IL6ST /CD130 can bind to IL6 /IL6R (alpha chain) complex, resulting in the formation of high-affinity IL6 binding sites, and transduce the signal. IL6ST /GP130 does not bind IL6 and may have a role in embryonic development.
