The coiled-coil domain is a structural motif found in proteins that are involved in a diverse array of biological functions such as the regulation of gene expression, cell division, membrane fusion and drug extrusion and delivery. CCDC34 (coiled-coil domain containing 34), also known as Renal carcinoma antigen NY-REN-41, is a 373 amino acid protein that is expressed in testis, breast, lung, placenta, liver and small intestine. A translocation between the short arms of chromosomes 11 and 18 affecting the CCDC34 gene has been identified in a patient with hamartoma of the retinal pigment epithelium. There are two isoforms of CCDC34 that are produced as a result of alternative splicing events.

FDPS is a 419 amino acid enzyme belonging to the FPP/GGPP synthetase family. Localized to cytoplasm and peroxisome, FDPS expression is regulated by phorbol esters and polyunsaturated fatty acids. FDPS assists in cholesterol biosynthesis, post-translational protein modifications and synthesis of steroid hormones in the isoprenoid pathway.FDPS catalyzes the formation of farnesyl diphosphate (FPP), a precursor for several classes of essential metabolites including sterols, dolichols, carotenoids, and ubiquinones. FDPS is inactivated by interferon-induced RSAD2, which may result in the disruption of lipid rafts at the plasma membrane. Existing as a homodimer, FDPS may have anti-viral effects when inactivated by RSAD2. Reduced activity of FDPS in liver may partly be the cause of Zellweger syndrome and neonatal adrenoleukodystrophy, both of which are known to be peroxisomal deficiency diseases.

Protein Kinase c alpha (PKC alpha) is an 77 kDa member of the conventional group (cPKCs: sensitive to calcium, diacylglycerol, phosphatidylserine and phorbol esters) of the PKC family of serine/ threonine kinases that are involved in a wide range of physiological processes including mitogenesis, cell survival and transcriptional regulation. PKC alpha is an ubiquitously expressed PKC isozyme that has been implicated in the regulation of a broad range of cellular functions including proliferation, differentiation, development, migration, cell cell adhesion, cell extracellular matrix adhesion, and solute transport. The activation loop threonine (threonine 497 in PKC alpha) of conventional PKCs is phosphorylated by phosphoinositide dependent kinase 1 (PDK1). This phosphorylation is necessary for the autophosphorylation of threonine 638 in the carboxy terminus of PKC alpha, a step that is critical for regulating the rate of PKC alpha dephosphorylation and inactivation.

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play distinct roles in cells. The protein encoded by this gene is one of the PKC family members. Studies both in human and mice demonstrate that this kinase is involved in B cell signaling and in the regulation of growth, apoptosis, and differentiation of a variety of cell types. Alternatively spliced transcript variants encoding the same protein have been observed. [provided by RefSeq, Jul 2008].
