APLF is a 511 amino acid protein that contains one FHA doman and two C2H2type zinc fingers. Localized to both the nucleus and the cytoplasm, APLF interacts with XRCC1, XRCC4 and Ku-86 and, via these interactions, is involved in single-strand and double-strand DNA break repair. APLF is subject to post-translational phosphorylation in response to DNA breaks. The gene encoding APLF maps to human chromosome 2, which houses over 1,400 genes and comprises nearly 8% of the human genome. Harlequin icthyosis, a rare and morbid skin deformity, is associated with mutations in the ABCA12 gene, while the lipid metabolic disorder sitosterolemia is associated with defects in the ABCG5 and ABCG8 genes. Additionally, an extremely rare recessive genetic disorder, is caused by mutations in the ALMS1 gene, which maps to chromosome 2.

Functions as a calcium permeable cation channel involved in fluid-flow mechanosensation by the primary cilium in renal epithelium. Together with TRPV4, forms mechano- and thermosensitive channels in cilium (PubMed:18695040). PKD1 and PKD2 may function through a common signaling pathway that is necessary for normal tubulogenesis. Acts as a regulator of cilium length, together with PKD1. The dynamic control of cilium length is essential in the regulation of mechanotransductive signaling. The cilium length response creates a negative feedback loop whereby fluid shear-mediated deflection of the primary cilium, which decreases intracellular cAMP, leads to cilium shortening and thus decreases flow-induced signaling. Also involved in left/right axis specification downstream of nodal flow: forms a complex with PKD1L1 in cilia to facilitate flow detection in left/right patterning (By similarity).

Spliceosomes are multi-protein complexes that are composed of snRNPs (small nuclear ribonucleoproteins) and a variety of associated protein factors, all of which work in concert to regulate the splicing of pre-mRNA, a critical step in the post-transcriptional regulation of gene expression. CWC22 (CWC22 spliceosome-associated protein), also known as NCM, fSAPb or EIF4GL, is a 908 amino acid nuclear protein and component of the spliceosome C complex. CWC22 is associated with the spliceosome prior to catalytic steps and remains associated throughout the reaction. Belonging to the CWC22 family, CW22 contains one MI domain and a MIF4G domain. The gene encoding CWC22 maps to human chromosome 2q31.3. As the second largest human chromosome, chromosome 2 makes up approximately 8% of the human genome and contains 237 million bases encoding over 1,400 genes.

BEGAIN is a 593 amino acid protein that localizes to cytoplasm and membrane. BEGAIN interacts with PSD-95 and SAPAP1 and forms a ternary complex and may sustain the structure of the postsynaptic density (PSD). BEGAIN is a novel PSD component associated with the core complex of PSD-95 and SAPAP. Because BEGAIN and SAPAP interact with the same region of PSD-95, BEGAIN and SAPAP may compete for the binding to PSD-95 and cannot interact with PSD-95 simultaneously. The C-terminal region of BEGAIN is involved in the interaction with PSD-95 whereas the N-terminal region has a coiled-coil structure that may interact with other molecules. BEGAIN is specifically expressed in brain and enriched in the PSD fraction. BEGAIN is also expressed in neurons and enriched at synaptic junctions, and is likely involved in the organization of synaptic junction components.

Rab proteins, a family of Ras-related small GTP-binding proteins, play a key role in regulating intracellular vesicle trafficking (1?). Rab GDP dissociation inhibitor (Rab GDI or GDI2) forms a soluble complex with Rab proteins and thereby prevents the exchange of GDP for GTP (1?). In mammals, there exist two major isoforms, Rab GDI alpha (also known as XAP-4) and Rab GDI beta (1,4). While the mammalian Rab GDI beta-genes are ubiquitously expressed, the Rab GDI ?genes are predominantly brain-specific (1). Since it is expressed predominantly in neural and sensory tissues, Rab GDI a may serve a specific function in neural signal transmission (5). The gene sequences for the Rab GDI proteins are extremely conserved in evolution, with substantial homology preserved across three eukaryotic kingdoms (5).

In mammalian cells, translation is controlled at the level of polypeptide chain initiation by initiation factors. Eukaryotic translation initiation factor 1 (eIF1) is crucial for the scanning process in vitro. During the scanning process, eIF1 is a component of a complex involved in recognition of the initiator codon. Translation is also initiated by the role of eIF1 in regulating the activity of ribosomal subunits 43S, 48S and 40S. eIF1 enables 43S ribosomal complexes to discern between cognate and near-cognate initiation codons, sensing the nucleotide content of initiation codons. It is also a promotor, along with eukar-yotic translation initiation factor 1A (eIF1A), for assembly of 48S ribosomal complexes at the initiation codon of a conventional capped mRNA. In addition, eIF1 and eIF1A, together with eukaryotic translation initiation factor 5 (eIF5), function in the formation of stable 40S ribosomal preinitiation complexes.

Dipeptidyl peptidases (DPPs) mediate regulatory activity of their substrates and have been linked to a variety of diseases including type 2 diabetes, obesity and cancer. DPPs have post-proline dipeptidyl aminopeptidase activity, cleaving Xaa-Pro dipeptides from the N-termini of proteins. DPPs can bind specific voltage-gated potassium channels and alter their expression and biophysical properties and may also influence T cells. DPP proteins include DPRP1, DPRP2, DPP3, DPP7, DPP10, DPPX and CD26. DPP3 (dipeptidyl-peptidase 3), also known as DPPIII, is a zinc-exopeptidase that belongs to the peptidase M49 family. DPP3 localizes to the cytoplasm and is involved in intracellular protein catabolism. More specifically, DPP3 is an important enzyme involved in the degradation of enkephalins. An increase in the activity of DPP3 is implicated in ovarian and endometrial cancers.

Dab1, a homolog of the Drosophila Disabled protein, is an adaptor protein involved in neural development. This cytoplasmic protein is tyrosine-phosphorylated during rapid expansion of the developing nervous system, and it is thought to interact with other proteins via a domain similar to the PTB domains of the Shc family. Dab1 has been shown to interact with the SH2 domains of Src, Fyn and Abl (1). Mutations in Dab1 result in widespread abnormalities in the brain, similar to those seen in Reelin mutants (2). Reelin is a secreted protein thought to play a role in directing migrating neurons (3). Evidence suggests that Dab1 functions downstream of Reelin in a signaling pathway involved in positioning cells in the developing brain (4). Dab2 (also designated DOC-2) is a mitogen-responsive phosphoprotein that binds the SH3 domain of Grb2, and it is thought to be a negative regulator of growth (5).

Calcium-dependent secretion activators (CAPS-1 and CAPS-2) are calcium-binding proteins that direct neurotransmitter and neuropeptide-filled vesicles to the cell membrane for secretory granule exocytosis. Both CAPS-1 and CAPS-2 are expressed primarily in the brain where they regulate the secretion of various substances. The CAPS proteins contain a PH domain that is essential for regulation of exocytosis, as well as regulation of phospholipid binding. Through their regulation of neurotrophin release from granule cells, CAPS proteins help to regulate cell fate during neuronal development. CAPS-1 is thought to regulate catecholamine release from neuronal cells, while CAPS-2 is thought to regulate release of both brain-derived neurotrophic factor and neurotrophin-3 from granule cells. Defects in the genes encoding CAPS-1 and CAPS-2 are implicated in impaired cerebral development and autism.

Transcription factor that plays a key role in male sex determination and differentiation by controlling testis development and male germ cell proliferation. Plays a central role in spermatogonia by inhibiting meiosis in undifferentiated spermatogonia and promoting mitosis, leading to spermatogonial development and allowing abundant and continuous production of sperm. Acts both as a transcription repressor and activator: prevents meiosis by restricting retinoic acid (RA)-dependent transcription and repressing STRA8 expression and promotes spermatogonial development by activating spermatogonial differentiation genes, such as SOHLH1. Also plays a key role in postnatal sex maintenance by maintaining testis determination and preventing feminization: represses transcription of female promoting genes such as FOXL2 and activates male-specific genes. May act as a tumor suppressor. May also play a minor role in oogenesis (By similarity).

Cell adhesion molecule that plays a role in neuronal self-avoidance. Promotes repulsion between specific neuronal processes of either the same cell or the same subtype of cells. Mediates within retinal amacrine and ganglion cell subtypes both isoneuronal self-avoidance for creating an orderly dendritic arborization and heteroneuronal self-avoidance to maintain the mosaic spacing between amacrine and ganglion cell bodies. Receptor for netrin required for axon guidance independently of and in collaboration with the receptor DCC. In spinal chord development plays a role in guiding commissural axons projection and pathfinding across the ventral midline to reach the floor plate upon ligand binding. Enhances netrin-induced phosphorylation of PAK1 and FYN. Mediates intracellular signaling by stimulating the activation of MAPK8 and MAP kinase p38.

Ubiquitination is an important mechanism through which three classes of enzymes act in concert to target short-lived or abnormal proteins for destruction. The three classes of enzymes involved in ubiquitination are the ubiquitin-activating enzymes (E1s), the ubiquitin-conjugating enzymes (E2s) and the ubiquitin-protein ligases (E3s). Ubr2 (Ubiquitin-protein ligase E3-alpha-2), also known as N-recognin-2, is a 1755 amino acid protein that contains one UBR-type zinc finger and one RING-type zinc finger. Participating in protein modification events within the N-end rule pathway, Ubr2 functions as an E3 ubiquitin-protein ligase that recognises and binds proteins that contain destabilising N-terminal residues, thereby leading to their ubiquitination and subsequent degradation. Mice lacking Ubr2 are infertile due to defects in male meiosis.

Existing as a heterodimer with Mms2 (also known as UBE2V2), UBC13 catalyzes the ATP-dependent synthesis of non-canonical polyubiquitin chains, a process that does not lead to proteasomal degradation. Additionally, UBC13 mediates the transcription of several target genes and is thought to play a role in cell cycle progression, cellular differentiation and DNA repair mechanisms that ensure cell survival after DNA damage.

Modulation of the chromatin structure plays an important role in the regulation of transcription in eukaryotes. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

Double-stranded RNA (dsRNA) endoribonuclease playing a central role in short dsRNA-mediated post-transcriptional gene silencing. Cleaves naturally occurring long dsRNAs and short hairpin pre-microRNAs (miRNA) into fragments of twenty-one to twenty-three nucleotides with 3' overhang of two nucleotides, producing respectively short interfering RNAs (siRNA) and mature microRNAs. SiRNAs and miRNAs serve as guide to direct the RNA-induced silencing complex (RISC) to complementary RNAs to degrade them or prevent their translation. Gene silencing mediated by siRNAs, also called RNA interference, controls the elimination of transcripts from mobile and repetitive DNA elements of the genome but also the degradation of exogenous RNA of viral origin for instance. The miRNA pathway on the other side is a mean to specifically regulate the expression of target genes.
