Voltage-gated K+ channels in the plasma membrane are important regulators of electrical Signalling, controlling the repolarisation and the frequency of action potentials in neurons, muscles and other excitable cells. KCNT2 is a 1,135 amino acid multi-pass transmembrane protein belonging to the potassium channel family (calcium-activated subfamily) of proteins. KCNT2 produces rapidly activating outward rectifier potassium currents in reponse to high intracellular sodium and chloride levels. Its channel activity is inhibited by ATP, inhalation anesthetics, such as isoflourane, and upon stimulation of G-protein coupled receptors, such as mAChR M1 and GluR-1. There are four isoforms of KCNT2 that are produced as a result of alternative splicing events.

OXSR1 is a serine/threonine kinase which regulates downstream kinases in response to environmental stress such as osmotic stresses, notably sorbitol and, to a lesser extent, NaCl. OXSR1 phosphorylated thr84 within the N-terminal regulatory domain of PAK1. Replacement of thr84 with gln reduced activation of PAK1 by an active form of the small G protein CDC42, suggesting that phosphorylation by OXSR1 modulates the G protein sensitivity of PAK. OXSR1 interacts with chloride channel proteins SLC12A6 isoform 2, SLC12A1 and SLC12A2 but not with SLC12A4 and SLC12A7, possibly establishing sensor/signaling modules that initiate the cellular response to environmental stress. Binds to and phosphorylates RELL1, RELL2 AND RELT. OXSR1 may have a role in regulating the actin cytoskeleton.

Band 3, also designated AE1, is an erythrocyte membrane glycoprotein that contributes to cell stuctural integrity and mediates exchange of chloride and bicarbonate across the phospholipid bilayer. The diverse functions of the approximately 900 amino acid protein are mediated by two distinct domains. The amino terminal domain, also known as cdb3 for cytoplasmic domain of erthrocyte membrane band 3, acts as an attachment site for the erythrocyte skeleton by binding ankyrin. The carboxy-terminal, membrane-associated domain carries out exchange transport of anions. Degradation of band 3 can generate an aging antigen known as senescent cell antigen, or SCA, which is expressed on old cells and marks them for removal by the immune system. An isoform of band 3, which lacks the first 65 amino acids and does not bind ankryin, is expressed in kidney.
