Beschreibung:   The annexin family of calcium-binding proteins is composed of at least ten mammalian genes. It is characterised by a conserved core domain, which binds to phospholipids in a Ca²⁺ dependent manner, and a unique amino terminal region, which may confer binding specificity. The annexin family has been implicated as regulators of such diverse processes as ion-flux, endocytosis and exocytosis, and cellular adhesion. Two forms of Annexin XI, designated A and B, have been identified. Transfection of COS-7 cells with Annexin XI-A, but not Annexin XI-B, causes formation of Annexin XI-associated vesicles.

Beschreibung:   In all vertebrates, SV2 proteins are abundant, hydrophobic, membrane glycoproteins that are expressed as two major isoforms, SV2A and SV2B, and one minor isoform, SV2C. SV2 proteins are differentially expressed in the brain and are present on all synaptic vesicles, independent of transmitter type. SV2A is abundantly expressed in the subcortex, specifically in the synaptic vesicles of all presynaptic nerve terminals, and also in most neuroendocrine secretory granules. SV2B displays a more restricted pattern of expression in that it is only present on a small subset of synapses in the hippocampus and cortex. SV2A and SV2B are functionally redundant and are required for maintaining normal brain function in vertebrates. SV2A and SV2B mediate synaptic transmission by regulating cytoplasmic Ca²⁺ levels in the nerve terminal during repetitive stimulation.

Beschreibung:   This gene encodes coagulation factor VIII, which participates in the intrinsic pathway of blood coagulation; factor VIII is a cofactor for factor IXa which, in the presence of Ca+2 and phospholipids, converts factor X to the activated form Xa. This gene produces two alternatively spliced transcripts. Transcript variant 1 encodes a large glycoprotein, isoform a, which circulates in plasma and associates with von Willebrand factor in a noncovalent complex. This protein undergoes multiple cleavage events. Transcript variant 2 encodes a putative small protein, isoform b, which consists primarily of the phospholipid binding domain of factor VIIIc. This binding domain is essential for coagulant activity. Defects in this gene results in hemophilia A, a common recessive X-linked coagulation disorder. [provided by RefSeq, Jul 2008].

Beschreibung:   The cadherins are a family of Ca++-dependent adhesion molecules that function to mediate cell-cell binding critical to the maintenance of tissue structure and morpho-genesis (1-4). Cadherins each contain a large extracellular domain at the amino terminus, which is characterized by a series of five homologous repeats, the most distal of which is thought to be responsible for binding specificity (5,6). The relatively short carboxy terminal, intracellular domain interacts with a variety of cytoplasmic proteins, including catenin b, to regulate cadherin function (7). BR-cadherin (for brain-cadherin, also designated cadherin-12 or N-cadherin 2) is expressed specifically in neurons in the central nervous system and is thought to be involved in neuronal development (8).

Beschreibung:   The product of this gene belongs to the Serine/Threonine protein kinase family, and to the Ca(2+)/calmodulin-dependent protein kinase subfamily. This protein plays a role in the calcium/calmodulin-dependent (CaM) kinase cascade by phosphorylating the downstream kinases CaMK1 and CaMK4. Seven transcript variants encoding six distinct isoforms have been identified for this gene. Additional splice variants have been described but their full-length nature has not been determined. The identified isoforms exhibit a distinct ability to undergo autophosphorylation and to phosphorylate the downstream kinases. [provided by RefSeq, Jul 2008].

Beschreibung:   The downstream of kinase family (Dok1-7) are members of a class of “docking” proteins that include the tyrosine kinase substrates IRS-1 and Cas, which contain multiple tyrosine residues and putative SH2 binding sites. Based on their similarities, the Dok family of proteins can be divided into three subgroups: Dok-1/2/3, Dok-4/5/6 and Dok-7. Through its interaction with muscle-specific receptor kinase (MuSK), Dok-7 is crucial for neuromuscular synaptogenesis and for MuSK activation. Mice lacking Dok-7 do not form neuromuscular synapses nor acetylcholine receptor clusters. Mutations in the Dok-7 gene can cause congenital myasthenic syndromes (CMA) — recessively inherited disorders characterized by muscle weakness.

Beschreibung:   Protein phosphatase 1 (PP1) is essential for cell division, and participates in the regulation of glycogen metabolism, muscle contractility and protein synthesis. The protein is involved in regulation of ionic conductances and long term synaptic plasticity. It may play an important role in dephosphorylating substrates such as the postsynaptic density associated Ca (2+)/calmodulin dependent protein kinase II.PP1 comprises a catalytic subunit, PPP1CA, PPP1CB or PPP1CC (PP1C gamma), which is folded into its native form by inhibitor 2 and glycogen synthetase kinase 3, and then complexed to one or several targeting or regulatory subunits. PPP1R12A and PPP1R12B mediate binding to myosin. PPP1R3A, PPP1R3B, PPP1R3C and PPP1R3D mediate binding to glycogen.

Beschreibung:   CAPS1 is a 189 amino acid cytoplasmic protein that contains four EF-hand domains, which serve as calcium-binding sites, and was first identified in canine thyroid. Interestingly, CAPS1 is much less abundant in humans than in canines. Synthesis and phosphorylation of CAPS1 is upregulated by cAMP-agonists in thyrocytes. CAPS1 likely functions in the regulation of ionic transport and may be involved in cross-signaling between cAMP and Ca(+2)-phophatidylinositol cascades. In addition to thyriod, CAPS1 is expressed in brain, salivary glands and lung. Expression of CAPS1 is increased in endometrial cancer and prognosis seems to be dependent on the level of CAPS1 expression, indicating that CAPS1 may be an appropriate prognostic marker for patient survival.

Beschreibung:   Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. This gene product belongs to a family of glutamate receptors that are sensitive to alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate(AMPA), and function as ligand-activated cation channels. These channels are assembled from 4 related subunits, Gria1-4. The subunit encoded by this gene (Gria2) is subject to RNA editing(CAG->CGG; Q->R) within the second transmembrane domain, which is thought to render the channel impermeable to Ca(2+). Alternative splicing, resulting in transcript variants encoding different isoforms, (including the flip and flop isoforms that vary in their signal transduction properties), has been noted for this gene.
