The protein encoded by this gene is a member of the Ser/Thr protein kinase family. This protein kinase is highly similar to the gene products of S. cerevisiae cdc28, and S. pombe cdc2. It is a catalytic subunit of the cyclin-dependent protein kinase complex, whose activity is restricted to the G1-S phase, and essential for cell cycle G1/S phase transition. This protein associates with and regulated by the regulatory subunits of the complex including cyclin A or E, CDK inhibitor p21Cip1 (CDKN1A) and p27Kip1 (CDKN1B). Its activity is also regulated by its protein phosphorylation. Two alternatively spliced variants and multiple transcription initiation sites of this gene have been reported. cdk2 is a cell cycle protein closely related to Cdc2 (cdk1) that has proved useful as a marker of proliferation. cdk2 binds cyclin type A and E proteins and controls progression into S-phase.

Centrosomes are dynamic organelles involved in many aspects of cell function and growth. Centrosomes act as microtubule organizing centers, and provide a site for concerted regulation of cell cycle progression. Duplication of centrosomes occurs once during each cell cycle and requires proper mitotic spindle formation and chromosome segregation. Defects in centrosome duplication or function are linked to many human diseases, including various forms of cancer. The centrosome and spindle pole-associated protein 1 (CSPP1) interacts with centrosomes and microtubules and may play a role in the regulation of G(1)/S-phase progression and spindle assembly. Two isoforms of CSPP1 exist as a result of alternative splicing events. Isoform 1 expression increases throughout the cell cycle and peaks in G2/M phase, whereas isoform 2 expression is highest in G1 phase and decreases thereafter.

DNA damage results in the arrest of cell cycle progression, allowing the damaged DNA to be repaired prior to replication. Checkpoints exist at several cell cycle phase transitions to maintain this genetic integrity. Rad9, Rad17, Rad24 and Mec3 are involved in activating the G1 and G2 checkpoints (1–4). Pol2 (also known as Dun2), encoding the catalytic subunit of DNA polymerase epsilon, plays a role in activating the S phase checkpoint (5). The protein kinase Rad53 (also designated Spk1, Mec2 or Sad1) is essential for both G2 and S phase arrest. Activation of Rad53 is regulated by Mec1 (also known as Esr1 and Sad3), a homolog of the human ATM protein (6). Pds1 and Mad2 both regulate checkpoints associated with incomplete spindle replication (7,8). Dun1, another protein kinase, plays a role in transducing the DNA damage signal (9).

Required for S phase entry of the cell cycle.The eukaryotic cell division cycle consists of a number of gene-controlled sequences that involve cyclin dependent kinases (Cdks) and cell division control (Cdc) proteins. Cdc123 (Cell division cycle protein 123), also known as D123, is a 336 amino acid cytoplasmic protein that is involved in cell cycle control. Widely expressed with high expression in thymus, spleen, ovary, testis, small intestine and leukocytes, Cdc123 functions to destabilize Chfr (checkpoint with forkhead and ring finger domain) proteins which, when accumulated, block the G to S phase transition. Cdc123 prevents the Chfr proteins from collecting in the cell, thereby allowing the cell to enter the S phase. Due to its role in cell cycle control, Cdc123 is thought to be a basal marker for luminal breast cancers.
