UCMA is a 138 amino acid secreted protein that is highly expressed in resting chrondrocytes in developing long bones and is thought to function in the early phase of chrondrocyte differentiation. A furin-like protease processes UCMA into an N-terminal 37 amino acid peptide and a C-terminal 74 amino acid peptide, which is referred to as Unique cartilage matrix-associated protein C-terminal fragment (Ucma-C). Introduction of recombinant Ucma-C interferes with osteogenic differentiation of mesenchymal stem cells, MC3T3-E1 preosteoblasts and primary osteoblasts. This suggests that Ucma may be involved in the negative regulation of osteogenic differentiation of osteochondrogenic precursor cells at the cartilage-bone interface and in peripheral zones of fetal cartilage.

The protein encoded by this gene is a member of the INK4 family of cyclin-dependent kinase inhibitors. This protein has been shown to form a stable complex with CDK4 or CDK6, and prevent the activation of the CDK kinases, thus function as a cell growth regulator that controls cell cycle G1 progression. The abundance of the transcript of this gene was found to oscillate in a cell-cycle dependent manner with the lowest expression at mid G1 and a maximal expression during S phase. The negative regulation of the cell cycle involved in this protein was shown to participate in repressing neuronal proliferation, as well as spermatogenesis. After dimerization they bind the RPA complex and are recruited to single stranded DNA. ATRIP is a nuclear protein that may also play a role in protein stabilization.

This gene encodes coagulation factor XI of the blood coagulation cascade. This protein is present in plasma as a zymogen, which is a unique plasma coagulation enzyme because it exists as a homodimer consisting of two identical polypeptide chains linked by disulfide bonds. During activation of the plasma factor XI, an internal peptide bond is cleaved by factor XIIa (or XII) in each of the two chains, resulting in activated factor XIa, a serine protease composed of two heavy and two light chains held together by disulfide bonds. This activated plasma factor XI triggers the middle phase of the intrisic pathway of blood coagulation by activating factor IX. Another family of proteins, Cdk inhibitors, also plays a role in regulating the cell cycle by binding to cyclin-Cdk complexes and modulating their activity. CDKL5 (cyclin-dependent kinase-like 5) is a 1030 amino acid protein that belongs to the CMGC Ser/Thr protein kinase family. Expressed in brain, lung, kidney, prostate, ovary, placenta, pancreas and testis, CDKL5 is thought to play a role in cell cycle regulation. Defects in CDKL5 are a cause of several disorders, such as X-linked infantile spasm syndrome and Rett syndrome.
