The constitutive enzyme glutathione reductase transforms glutathione into its reduced state which ultimately can provide a measure of cellular toxicity. GSTT2 (glutathione S-transferase theta-2), also known as GST class-theta-2, is a 244 amino acid enzyme with sulfatase activity that functions in conjugating reduced glutathione to hydrophobic electrophiles. GSTT2 exists as a homodimer in the cytoplasm and is expressed in low levels in the liver and the lung. GSTT2 belongs to the GST superfamily and contains both a GST C-terminal and a GST N-terminal domain. The gene encoding GSTT2 exists on human chromosome 22.

This enzyme also plays a significant role in the catabolism of phenylalanine and tyrosine. Several transcript variants of this gene encode multiple protein isoforms. GSTZ1 shows minimal glutathione-conjugating activity with ethacrynic acid and 7-chloro-4-nitrobenz-2-oxa-1,3-diazole and maleylacetoacetate isomerase activity. It has low glutathione peroxidase activity with T butyl and cumene hydroperoxides and is able to catalyze the glutathione dependent oxygenation of dichloroacetic acid to glyoxylic acid. Highest expression in liver followed by kidney, skeletal muscle and brain. Also expressed in melanocytes, synovium, placenta, breast and fetal liver and heart.

GSTCD (glutathione S-transferase, C-terminal domain containing) is a 633 amino acid protein belonging to the GSTCD family and contains one GST C-terminal domain. The gene encoding GSTCD maps to human chromosome 4, which represents approximately 6% of the human genome and contains nearly 900 genes. Notably, the Huntingtin gene, which is found to encode an expanded glutamine tract in cases of Huntington's disease, is on chromosome 4. FGFR-3 is also encoded on chromosome 4 and has been associated with thanatophoric dwarfism, achondroplasia, Muenke syndrome and bladder cancer. Chromosome 4 is also tied to Ellis-van Creveld syndrome, methylmalonic acidemia and polycystic kidney disease.
