Glycogen synthesis is initiated by the autoglucosylation of Glycogenin-1. Specifically, Glycogenin-1 glucosylates itself to begin the synthesis of glycogen in mammalian skeletal muscle. It acts as the primer to which further glucose monomers may be added. All of the Glycogenin-1 molecules contain at least one glucosyl residue before autoglucosylation begins. The first step of the glycogen synthesis occurs when a glucose molecule from UDP-glucose binds to the hydroxyl group of Tyr 194 on the Glycogenin-1 molecule. Using its glucosyltransferase activity, Glycogenin-1 adds more glucoses, each one coming from UDP-glucose. The glycosylation process reaches a plateau when five new glucose residues have been added, at which point glycogen synthase (GS) takes over and further elongates the chain. Glycogenin-1 remains covalently attached to the reducing end of the glycogen molecule.

The GAT1 gene family includes sodium- and chloride-dependent plasma membrane transporters for neurotransmitters, metabolites and osmolites, which couple substrate flux to transmembrane electrochemical gradients. PROT (Sodium-dependent proline transporter), also known as Solute carrier family 6 member 7, is a 636 amino acid multi-pass membrane protein that is a GAT1 family member specifically expressed in regions of the brain. PROT terminates the action of proline by its high affinity sodium/chloride-dependent reuptake into pre-synaptic terminals. Enriched in glutamatergic synaptic terminals, it is likely that PROT plays an important role in excitatory events of neurotransmission. PROT-mediated proline uptake is inhibited by compounds such as benztropine, LP-403812 and Des-Tyr-Leu-enkephalin (GGFL). These inhibitors of proline uptake may lead to the development of therapeutic agents for certain neurologic disorders.

p44/42 MAP Kinase(Thr202); ERK (extracellular signal regulated kinase), also known as MAPK (mitogen activated protein kinase) has two closely related isoforms of 44 kDa and 42 kDa, respectively. These kinases belong to a family of serine/threonine kinases that are activated upon treatment of cells with a large variety of stimuli including mitogens, hormones, growth factors, cytokines, and bioactive peptides. Cell stimulation induces the activation of a signaling cascade, the downstream effects of which have been linked to the regulation of cell growth and differentiation as well as the cytoskeleton. ERK1 and ERK2 are phosphorylated within the activation loop on both a Threonine and a Tyrosine residue (within a Thr-Glu-Tyr motif) by MEKs (MAPK/ERK kinases), thereby greatly elevating the activity of ERK1&2.
