GLYCTK is a 523 amino acid protein that is expressed as seven isoforms which are present throughout the body. Localized to the cytoplasm and the mitochondrion in an isoform-specific manner, GLYCTK functions to catalyze the ATP-dependent conversion of (R)-glycerate to 3-(R)-glycerate, thereby playing an important role in neural and skeletal muscle systems. Defects in the gene encoding GLYCTK are the cause of D-glyceric acidemia, an inborn error of amino acid metabolism that is best described as nonketotic hyperglycinemia and is characterized by the excretion of D-glyceric acid in the urine.

Mouse NGF (2.5S) was isolated from mouse submaxillary glands by method of Mobley et al (1976) and is a form of beta-NGF that has identical biological properties. NGF is known to regulate the survival and development of certain sympathetic and sensory neurons. It is a dimer with 2 identical polypeptide chains and dimeric molecular weight of approximately 26,500 Da. Isolation and purification of NGF from mouse submaxillary glands yields preparations of NGF (2.5S) with identical biological activity but with cleavages at the amino terminus (with the loss of 8 amino acids) and/or at the carboxy-terminus (with the loss of arginine). These preparations are named 2.5 NGF.

Required for the function of light chain amino-acid transporters. Involved in sodium-independent, high-affinity transport of large neutral amino acids such as phenylalanine, tyrosine, leucine, arginine and tryptophan. Involved in guiding and targeting of LAT1 and LAT2 to the plasma membrane. When associated with SLC7A6 or SLC7A7 acts as an arginine/glutamine exchanger, following an antiport mechanism for amino acid transport, influencing arginine release in exchange for extracellular amino acids. Plays a role in nitric oxide synthesis in human umbilical vein endothelial cells (HUVECs) via transport of L-arginine. Required for normal and neoplastic cell growth. When associated with SLC7A5/LAT1, is also involved in the transport of L-DOPA across the blood-brain barrier, and that of thyroid hormones triiodothyronine (T3) and thyroxine (T4) across the cell membrane in tissues such as placenta. When associated with SLC7A5 or SLC7A8, involved in the cellular activity of small molecular weight nitrosothiols, via the stereoselective transport of L-nitrosocysteine (L-CNSO) across the transmembrane. Together with ICAM1, regulates the transport activity LAT2 in polarised intestinal cells, by generating and delivering intracellular signals. When associated with SLC7A5, plays an important role in transporting L-leucine from the circulating blood to the retina across the inner blood-retinal barrier.

Expressed predominately in forebrain tissue, LRRTM1 is thought to be involved in the development of forebrain structures, specifically by influencing axon trafficking, as well as neuronal differentiation and connectivity. Human LRRTM1 shares 96% amino acid identity with its mouse counterpart, suggesting a conserved role between species. Defects in the gene encoding LRRTM1 may be associated with the pathogenesis of several common neurodevelopmental disorders.
