Studies of the orthologous gene in mouse indicate that it regulates late endosomal traffic and cell proliferation. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. A pseudogene of this gene is located on the long arm of chromosome 13. Cytochrome c oxidase subunit I (COI or MTCO1) is one of three mitochondrial DNA (mtDNA) encoded subunits (MTCO1, MTCO2, MTCO3) of respiratory Complex IV. Complex IV is located within the mitochondrial inner membrane and is the third and final enzyme of the electron transport chain of mitochondrial oxidative phosphorylation. Complex IV is composed of 13 polypeptides. Subunits I, II, and III (MTCO1, MTCO2, MTCO3) are encoded by mtDNA while subunits IV, Va, Vb, VIa, VIb, VIc, VIIa, VIIb, VIIc, and VIII are nuclear encoded. Mammalian MTCO1 has 12 membrane-spanning alpha-helices (I to XII).

Synaptotagmins (Syts) are a large gene family of synaptic vesicle integral membrane proteins. Syt functions as regulators of both exocytosis /endocytosis and is involved in neurotransmitter secretion from small secretory vesicles. 13 isoforms of Syt have been identified(SytI-XIII). SynaptotagminVI is localised to the endoplasmic reticulum and/or Golgi like perinuclear compartment. It may be important for trafficking and calcium signaling as it is specially expressed in nonneuronal tissues. Also SytVI is present in the acrosomal region of mammalian spermatozoa. The cytosolic domain of SytVI can abrogate exocytosis by competing with the endogenous protein for essential interactions with the fusion machinery involved in a acrosomal exocytosis.

Hemogen is a 484 amino acid protein encoded by the human gene HEMGN. Hemogen is a nuclear protein that is expressed in hematopoietic precursor cells and can be detected in CD34+ and K-562 leukemia cell line. It is also expressed in bone marrow, testis, thymus and thyroid tumors, non-Hodgkin lymphoma, various leukemia cell lines, peripheral blood mononuclear cells (PBMCs) and bone marrow mononuclear cells (BMMCs) of patients with leukemia. Hemogen is down-regulated during megakaryocytic differentiation of K-562 cells by 12-O-tetradecanoylphorbol-13-acetate (TPA) (at protein level). It can be up-regulated in normal PBMCs by mitogens.

The complex consists of 13 mitochondrial- and nuclear-encoded subunits. The mitochondrially-encoded subunits perform the electron transfer of proton pumping activities. The functions of the nuclear-encoded subunits are unknown but they may play a role in the regulation and assembly of the complex. GBGT1 is a 347 amino acid single-pass type II membrane protein that belongs to the glycosyltransferase 6 family. Localizing to the golgi apparatus membrane, GBGT1 is widely expressed, with high levels found in placenta, ovary and peripheral blood leukocyte, and lower levels expressed in liver, thymus and testis. GBGT1 utilizes manganese as a cofactor, and assists in the addition of N-acetylgalactosamine (GalNAc) in alpha-1,3-linkage to various substrates, resulting in the formation of glycolipids. Glycolipids are present in most eukaryotic cells and may assist in the adherence of certain pathogens. Existing as two alternatively spliced isoforms, the gene encoding GBGT1 maps to human chromosome 9q34.2 and mouse chromosome 2 A3.
