SIRT7 is a human member of a family of proteins called Sirtuins (Sir2-like proteins) and are present in prokaryotes and eukaryotes. All Sir2-like proteins have a sirtuin core domain, which contains a series of sequence motifs conserved in organisms ranging from bacteria to humans. Bacterial, yeast and mammalian sirtuins are able to metabolize NAD and possibly at as mono-ADP-ribosyltransferases. The enzymatic function of sirtuins is not yet completely understood but recent reports of histone-activated Sir2-mediated NAD metabolism and NAD-activated Sir2-mediated histone deacetylation suggest a possible coupled reciprocal activation mechanism involving interactions of Sir2 with NAD and the N epsilon-acetyl-lysine groups of acetylated histones.

Als2Cr2, also called STE20-related kinase adapter protein beta (STRAD beta) and ILPIP, is a serine/threonine protein kinase encoded by the STRADB gene, discovered as an XIAP interacting protein. It is a mediator of the XIAP/TAK1 activation of the JNK1 pathway, an apoptosis inhibition mechanism independent of XIAP inhibition of caspases. Alone, it only moderately activates JNKs, but its interaction with XIAP/TAK1 induces a much stronger activation of JNK1, protecting against Caspase 1- or Fas-induced apoptosis.

Mesencephalic astrocyte-derived neurotrophic factor(MANF) is a secreted protein which belongs to the ARMET family. MANF selectively promotes the survival of dopaminergic neurons of the ventral mid-brain. It modulates GABAergic transmission to the dopaminergic neurons of the substantia nigra. MANF enhances spontaneous, as well as evoked, GABAergic inhibitory postsynaptic currents in dopaminergic neurons. MANF inhibits cell proliferation and endoplasmic reticulum (ER) stress-induced cell death. The N-terminal region of ARMET may be responsible for neurotrophic activity while the C-terminal region may play a role in the ER stress response. MANF reduces endoplasmic reticulum (ER) stress and has neurotrophic effects on dopaminergic neurons. Intracortical delivery of recombinant MANF protein protects tissue from ischemic brain injury. MANF has been described as a survival factor for dopaminergic neurons. MANF and a homologous protein, the conserved dopamine neurotrophic factor (CDNF), form a novel evolutionary conserved family of neurotrophic factors. MANF expression was widespread in the nervous system and non-neuronal tissues.

CARD8 Antibody: Apoptosis is related to many diseases and development. Cell death signals are transduced by death domain (DD), death effector domain (DED), and caspase recruitment domain (CARD) containing molecules. CARD containing proteins include some caspases, Apaf-1, CARD4, IAPs, RICK, ARC, RAIDD, BCL-10, and ASC. A novel CARD-containing protein was recently identified and designated CARD8. This protein interacts with DRAL, a p53-responsive protein implicated in the induction of apoptosis, and caspase-1 and its related proteins ICEBERG and pesudo-ICE. Although there are conflicting reports on whether CARD8 acts a pro- or anti-apoptotic protein, it has been suggested that it functions as an adaptor molecule regulating caspase-1 and NF-kappa B activation.

AIF Antibody: Apoptosis is characterized by several morphological nuclear changes including chromatin condensation and nuclear fragmentation. These changes are triggered by the activation of members of caspase family, caspase activated DNase, and several novel proteins. A novel gene, the product of which causes chromatin condensation and DNA fragmentation, was recently identified, cloned, and designated apoptosis inducing factor (AIF). Like the critical molecules, cytochrome c and caspase-9, in apoptosis, AIF localizes in mitochondria. AIF translocates to the nucleus when apoptosis is induced and induces mitochondria to release the apoptogenic proteins cytochrome c and caspase-9. AIF induces chromatin condensation and large scale DNA fragmentation, which are the hallmarks of apoptosis, of the isolated nucleus and the nucleus in live cells by microinjection and apoptosis stimuli. AIF is highly conserved between human and mouse and widely expressed.
