SPARC (secreted protein acidic and rich in cysteine) is a phosphorylated, acidic, glycine-rich glycoprotein that is secreted by endothelial cells and is present in large amounts in the parietal endoderm of mouse embryos and in human placenta. SPARC-like protein 1 (SPARCL1), also known as high endothelial venule protein (Hevin) or MAST9, is a 664 amino acid member of the SPARC family of proteins. Highly expressed in lymph node, heart, lung, brain, skeletal muscle, ovary, colon and small intestine, SPARCL1 is a secreted protein that contains one EF-hand domain, one follistatin-like domain and one Kazal-like domain. SPARCL1 is implicated to play a role in neuronal remodeling and tumor suppression. The gene encoding SPARCL1 maps to chromosome 4q22.1.

AIP1 (ASK-interacting protein 1), also known as DAB2IP (DAB2 interacting protein), is a 1189 amino acid protein that localizes to both the membrane and the cytoplasm and contains one Ras-GAP domain, one PH domain and one C2 domain. Expressed at low levels in prostate tissue, AIP1 functions as a Ras GTPase-activating protein that interacts with ASK 1 and, via this interaction, disrupts the association of ASK 1 with an inhibitory 14-3-3 complex, thereby allowing free ASK 1 to function within the cell. AIP1 exists as multiple alternatively spliced isoforms and is down-regulated in prostate cancer tissue, suggesting a role in tumour suppression. Chromosomal aberrations in the gene encoding AIP1 are associated with acute myeloid leukaemia (AML), implicating a role for AIP1 fusion proteins in tumourigenesis.

HES7, along with family member HES1, is thought to cooperatively regulate somite formation in the presomitic mesoderm. HES7 may also be essential for coordinated somite segmentation by acting as a segmentation clock. HES7 contains one basic helix-loop-helix (bHLH) domain and one Orange domain. Mutations in HES7 have been found to cause spondylocostal dysostosis, an autosomal recessive disorder characterized by deformities of the chest and spine.
