Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamate-gated, cation-specific ion channels. Kainate/AMPA receptors are co-localized with NMDA receptors in many synapses and consist of seven structurally related subunits designated GluR-1 to -7. The kainate/AMPA receptors are primarily responsible for the fast excitatory neuro-transmission by glutamate whereas the NMDA receptors are functionally characterized by a slow kinetic and a high permeability for Ca2+ ions. The NMDA receptors consist of five subunits: epsilion 1, 2, 3, 4 and one zeta subunit. The zeta subunit is expressed throughout the brainstem whereas the four epsilon subunits display limited distribution.

RNF185 (ring finger protein 185), also known as FLJ38628, is a 192 amino acid multi-pass membrane protein containing one RING-type zinc finger. Two RNF185 isoforms exist as a result of alternative splicing, and the gene encoding RNF185 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

ADAMTS proteases are secreted enzymes containing a prometalloprotease domain of the reprolysin type. The ADAMTS proteases function in processing of procollagens and von Willebrand factor as well as catabolism of aggrecan, versican and brevican. They have been demonstrated to have important roles in connective tissue organization, coagulation, inflammation, arthritis, angiogenesis and cell migration.A member of the metalloproteinase family containing disintegrin like domains (ADAMs), the function of ADAMTS8 is still poorly understood. ADAMTS8 contains the canonical HExxHxxxxxH zinc metalloproteinase motif, and has been shown to be proteolytically active on a range of substrates. ADAMTS8 is inhibited by the endogenous MMP inhibitors, TIMP1, 2, 3 and 4, but most efficiently by TIMP3. In addition to the metalloprotease domain, ADAMTS8 has a propeptide domain, a Prohormone Convertase (PC, furin) cleavage site, a cysteine rich domain and thrombospondin 1 like domains.

Regulation of neuronal synaptic adhesion by CAMs has proven important for learning and memory. Proper embryonic morphogenic development is also heavily dependent on the regulation of cell adhesion molecules. Neurotrimin (hNT) is a neural cell adhesion molecule localizing to the cell membrane, where it acts as a lipid-anchor. Neurotrimin belongs to the IgLON family of proteins, a member of the larger immunoglobulin superfamily.

Via its ability to regulate protein folding, TorsinB plays a key role in postnatal developmental events and is essential for proper neurological development. PTEN is an important tumor suppressor gene located on chromosome 10 and, when defective, causes a genetic predisposition to cancer development known as Cowden syndrome. The chromosome 10 encoded gene ERCC6 is important for DNA repair and is linked to Cockayne syndrome which is characterised by extreme photosensitivity and premature aging. Tetrahydrobiopterin deficiency and a number of syndromes involving defective skull and facial bone fusion are also linked to chromosome 10. As with most trisomies, trisomy 10 is rare and is deleterious.

Lissencephaly (smooth brain) is an abnormality of brain development characterized by incomplete neuronal migration and a smooth cerebral surface, manifesting as severe mental retardation. Genetic analysis has identified two proteins that are mutated in some cases of lissencephaly, designated lissencephaly-1 protein (LIS1) and doublecortin. LIS1 displays sequence homology to ?subunits of heterotrimeric G proteins, and doublecortin contains a consensus Abl phosphorylation site. In addition, the DCAMKL1 (doublecortin-like and CAM kinase-like 1) protein shows homology to doublecortin. All three proteins are highly expressed in developing brain and may function together to regulate microtubules involved in neuronal migration. The DCAMKL1 protein encodes a functional kinase that is capable of phosphorylating myelin basic protein and itself, but its kinase activity does not appear to affect its microtubule polymerization activity.

The alpha subunit of TSH is also present in two other pituitary glycoprotein hormones, follicle stimulating hormone and luteinizing hormone, and, in primates, in the placental hormone chorionic gonadotropin. Each of these hormones also has a unique beta subunit, which provides receptor specificity. In other words, TSH is composed of alpha subunit bound to the TSH beta subunit, and TSH associates only with its own receptor. Free alpha and beta subunits have essentially no biological activity.

PHYHIP (Phytanoyl-CoA hydroxylase-interacting protein) is a 330 amino acid protein that is strongly expressed in brain, with weak expression in ovary, small intestine and ovary. In transgenic mice, overexpression of PHYHIP in heart results in tachycardia and tachyarrhythmia. PHYHIP interacts with the Refsum disease gene product, PAHX, indicating that PHYHIP may play a role in the CNS deficits of Refsum disease, which is characterized by cerebellar degeneration, neurologic damage and peripheral neuropathies. PHYHIP also interacts with Dyrk1A, a protein that that is overexpressed in brain of Down-syndrome patients, therefore PHYHIP may participate in some of the neurological abnormalities of Down syndrome. Significantly, the gene encoding PHYHIP is localized to a region of the short arm of human chromosome 8 that is frequently found deleted in prostate, breast and several other types of cancers.

P450 enzymes constitute a family of monooxygenase enzymes that are involved in the metabolism of a wide array of endogenous and xenobiotic compounds (1). Several P450 enzymes have been classified by sequence similarities as members of the CYP1A and CYP2A subfamilies (2). NADPH cytochrome P450 reductase is a microsomal enzyme responsible for the transfer of electrons from NADPH to cytochrome P450 enzymes during the P450 catalytic cycle (3,4). NADPH cytochrome P450 reductase is localized to the endoplasmic reticulum where it is also able to transfer electrons to heme oxygenase and cytochrome b5 (5,6). NADPH cytochrome P450 reductase is structurally related to two separate flavoprotein families, ferredoxin nucleotide reductase (FNR) and flavodoxin (7). Electron transfer of NADPH cytochrome P450 reductase requires the binding of two flavin cofactors, FAD and FMN, to the FNR and flavodoxin domains, respectively (8).

As a subfamily of the cadherin superfamily, protocadherins are cadherin-like cell adhesion proteins that contain up to seven extracellular domains and are predominantly expressed in the nervous system. Importantly, the adhesion mechanism of protocadherins is distinct from classic cadherins. Through inactivation or overexpression, several protocadherins have been implicated in a variety of cancers. Protocadherin-20 (PCDH20), also known as protocadherin-13, is a 924 amino acid protein containing 6 cadherin domains and potentially functioning as a calcium-dependent cell-adhesion protein. In non-small cell lung cancer cell lines, a homozygous loss of PCDH20 was identified through either deletion of one allele and methylation of the other or methylation of both alleles. Hypermethylation of PCDH20 is associated with worse prognosis and clinical outcome, suggesting that PCDH20 may function as a tumor suppressor.
