Extracellular proteases mediate the digestion of neighboring extracellular matrix components in initial tumour growth, allow desquamation of tumour cells into the surrounding environment, provide the basis for invasion of basement membranes in targeted metastatic organs and are required for release and activation of many growth and angiogenic factors. TMPRSS5 (transmembrane protease, serine 5), also known as spinesin, is a 457 amino acid single-pass type II membrane protein that is expressed specifically in brain and is thought to play a role in hearing. A member of the peptidase S1 family, TMPRSS5 contains one peptidase S1 domain and an SRCR domain, and is encoded by a gene that maps to human chromosome 11q23.2. Defects in the gene encoding TMPRSS5 are associated with deafness.

EF-1 (elongation factor-1) is a multi-protein complex that is responsible for the delivery of aminoacyl-tRNAs to the ribosome. EF-1 gamma (elongation factor 1-gamma), also known as EEF1G or GIG35, is a 437 amino acid subunit of the EF-1 complex. Expressed in stomach, pancreas, brain, lung, kidney, intestine, liver and spleen, EF-1 gamma contains an N-terminal glutathione transferase domain which is thought to be involved in anchoring the complex to various cellular components. Additionally, EF-1 gamma may play a key role in the assembly of multiprotein complexes containing aminoacyl-tRNA synthetases. Increased expression of EF-1 gamma is associated with pancreatic cancer, suggesting a possible role for EF-1 gamma in the oncogenic transformation process.

ATP13A2 is a 1,180 amino acid multi-pass membrane protein that belongs to the P5 subfamily of ATPases which play an important role in the transportation of inorganic cations. Expressed as multiple alternative spliced isoforms, ATP13A2 functions to catalyze the conversion of ATP to ADP and a free phosphate, thereby participating in the active transport of ions across cellular membranes. Defects in the gene encoding ATP13A2 are the cause of Kufor-Rakeb syndrome (KRS), a rare hereditary type of Parkinson’s disease that exhibits juvenile onset and is characterized by neurodegeneration and dementia. The ATP13A2 gene maps to human chromosome 1, which spans 260 million base pairs, contains over 3,000 genes and comprises nearly 8% of the human genome.

NOB1P is a 412 amino acid nuclear protein that is involved in proteosome biogenesis and is required for the final step in 18S rRNA maturation. NOB1P contains a PIN domain, which functions as a nuclease in nonsense-mediated mRNA decay and is required for pre-rRNA cleavage. NOB1P interacts with Rent2, which is involved in nonsense-mediated decay of mRNAs containing premature stop codons. Expressed in placenta, spleen, endothelial cells, liver and lung, NOB1P is essential for the synthesis of 40S ribosome subunits. Supression of the gene encoding NOB1P inhibits the processing of the 20S pre-rRNA to the mature 18S rRNA, therefore leading to accumulation of high levels of 20S pre-rRNA with degradation intermediates.

ADH6 (alcohol dehydrogenase 6), also known as ADH-5, is a 368 amino acid member of the class V zinc-containing alcohol dehydrogenase family. This family of enzymes functions to metabolize a wide variety of substrates such as retinol, hydroxysteroids, ethanol, aliphatic alcohols and lipid peroxidation products. Localized to the cytoplasm and expressed in the stomach and liver, ADH6 catalyzes the reversible oxidation of alcohols to their corresponding aldehydes or ketones and is able to bind two zinc ions as cofactors. ADH6 contains a glucocorticoid response element upstream of its 5' UTR which is thought to be a steroid binding site, suggesting that expression of ADH6 may be under hormonal control. Multiple isoforms of ADH6 exist due to alternative splicing events.

Arylsulfatase F, also known as ARSF, is a 590 amino acid secretory protein that belongs to the sulfatase family of bone and cartilage matrix proteins. Arylsulfatase F uses calcium as a cofactor to catalyze reactions that are important in maintaining correct bone composition. The activity of Arylsulfatase F, unlike that of other family members, such as Arylsulfatase E, is not inhibited by warfarin. The gene encoding Arylsulfatase F maps to human chromosome X, which contains nearly 153 million base pairs and houses over 1,000 genes. In conjunction with chromosome Y, chromosome X is responsible for sex determination. There are a number of conditions related to an abnormal number and combination of sex chromosomes, some of which include Turner's syndrome, color blindness, hemophilia and Duchenne muscular dystrophy.

The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. RNF215 (ring finger protein 215), is a 377 amino acid multi-pass membrane protein containing one RING-type zinc finger. The gene encoding RNF215 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

CALHM1 is a 346 amino acid multi-pass endoplasmic reticulum membrane protein that belongs to the FAM26 family. CALHM1 co-localizes with GRP 78 to the endoplasmic reticulum. Predominantly expressed in adult brain, CALHM1 may be a pore-forming ion channel that controls cytosolic Ca2+ permeability and cytosolic Ca2+ concentration in the cell. It is suggested that CALHM1 regulates amyloid precursor protein proteolysis and aggregated amyloid-beta peptides levels in a Ca2+ dependent manner. CALHM1 homomultimerizes and shares strong sequence similarities with the selectivity filter of the NMDA receptor, which generates a large Ca2+ conductance across the plasma membrane. CALHM1 may be a potential factor involved in the pathogenesis of Alzheimer's disease.

EMP-2 is a 167 amino acid multi-pass membrane protein that contains four-transmembrane domains and belongs to the GAS3/PMP22 (growth arrest-specific-3/peripheral myelin protein-22) family. Localized to lipid raft domains in the plasma membrane, EMP-2 regulates the expression of several target proteins and is necessary for blastocyst implantation in the uterine endometrium. Specifically, EMP-2 mediates blastocyst implantation by controlling the cell membrane expression of MHC and glycosylphosphatidylinositol-anchored proteins, as well as Integrins and caveolin-1. In adult tissues, EMP-2 is expressed in heart, lung, ovary and intestine, while fetal expression is highest in kidney, brain and liver. Overexpression of EMP-2 is associated with endometrial adenocarcinoma, suggesting a possible role for EMP-2 in tumorigenesis.

YKL-39 is a 390 amino acid cartilage protein that belongs to the chitinase family of chitin-fragmenting hydrolases. Highly expressed in chondrocytes (cartilage cells) and synoviocytes (fibroblastic cells that line joint cavities), YKL-39 binds glycan structures with high affinity. Although related to bacterial chitinases, YKL-39 lacks the characteristic glutamate active site and, thus, does not have enzymatic chitinase activity. Patients affected with rheumatoid arthritis (RA) have autoimmunity against YKL-39, suggesting that YKL-39 is involved in osteoarthritic and/or rheumatoid joint disease. Additionally, YKL-39 is upregulated in early degenerative cartilage diseases (such as RA) and may be a marker of chondrocyte activation in these autoimmune conditions.

Prenylation and methylation are two forms of protein modification, both of which are important for a variety of functions, including membrane attachment, protein-protein interactions and Signalling events. NARF (nuclear prelamin A recognition factor), also known as IOP2, is a 456 amino acid nuclear protein that belongs to the NARF family. Expressed ubiquitously with highest expression in heart, skeletal muscle and brain, NARF binds to the C-terminal end of prenylated prelamin A and may be a member of a prelamin A-containing endoprotease complex. Additionally, via its association with prelamin A, NARF may be involved in heterochromatin organization. NARF is expressed as three isoforms due to alternative splicing events and, upon DNA damage, may be phosphorylated by ATM or ATR.

Dyskerin (NAP57) associates with the chaperone protein Nopp140 and forms a small ribonucleoprotein particle with GAR1 (NOLA1), NHP2 (NOLA2) and Nop10 for the isomerization of uridine to pseudouridine (1). GAR1, NHP2 and dyskerin localize to the dense fibrillar component of the nucleolus and in nuclear Cajal bodies (2). The dyskerin gene maps to chromosome Xq28 (3). Missense mutations in the dyskerin gene interfere with normal nuclear localization of dyskerin and cause Dyskeratosis congenita (DKC) (4). DKC is a rare, X-linked bone marrow disorder characterized by cutaneous hyperpigmentation, dystrophy of the nails, atrophy of the testicles and leukoplakia of the oral mucosa. The GAR1 gene maps to chromosome 4q25 (5,6). The NHP2 gene maps to chromosome 5q35.3 and encodes a 155-amino acid protein (2,7).
