GRASP (GRP1-associated scaffold protein, tamalin) is a 395 amino acid proteinencoded by the human gene GRASP. GRASP is a scaffold protein that com-prises multiple protein-interacting domains, including a 95 kDa postsynapticdensity protein (PSD-95)/discs-large/ZO-1 (PDZ) domain, a leucine-zipperregion and a carboxyl-terminal PDZ-binding motif. GRASP is involved withintracellular trafficking and contributes to the macromolecular organizationof group 1 metabotropic glutamate receptors (mGluRs) at synapses. GRASPforms a heteromer composed of GRASP , PSCD2 and at least one mGluR-1. Italso interacts with PSCD3, mGluR-2, mGluR-3 and mGluR-5. Tris have been useful as buffers in a wide variety of biological systems. It has been used as a starting material for polymers, oxazolones (with carboxylic acids) and oxazolidines (with aldehydes). It does not precipitate calcium salts and is of value in maintaining solubility of manganese salts. It can be used for the direct standardization of a strong acid solution; the equivalence point can be determined either potentiometrically or by use of a suitable indicator such as 3-(4-Dimethylamino-1-naphthylazo)-4-methoxybenzenesulfonic acid. It is RNAse and DNAse-free. Tris is relatively non-hygroscopic ; but, if needed, it can be dried at 100 °C for up to 4 hours to remove any water.
Tris is used in pH control in vitro and in vivo for body fluids and in buffering systems for electrophoresis applications.Tris is used in assays used to characterize the activity and kinetics of the enzymes that catalyze SUMOylation of Small ubiquitin-like proteins (SUMO) and SUMO-dependent protein-protein interactions.

D-Biotin is a growth factor present in small amounts in every living cell. It is involved in naturally occurring carboxylation reactions. It occurs mainly bound to proteins or polypeptides. The PDPH is a heterobifunctional crosslinker containing sulfhydryl-reactive pyridyldithiol and carbonyl-reactive hydrazide moieties. Pyridyldithiols react with free sulfhydryls (-SH) to form disulfide bonds. Hydrazide groups react with carbonyls (aldehydes and ketones) to form stable hydrazone bonds. Aldehyde groups can be created by periodate-oxidation of sialic acid and other sugar components of glycoprotein polysaccharides. Thus, PDPH is useful for conjugating glycoproteins and sulfhydryl-containing peptides or proteins. Likewise, PDPH is useful as a sulfhydryl-addition reagent for glycoproteins and other carbohydrates; after reaction of the hydrazide to an oxidized carbohydrate, the pyridyldithiol group can be cleaved by a reducing agent to expose a sulfhydryl group. Yet another application for PDPH is to react the primary amine of the hydrazide moiety to a carboxyl group using the crosslinker EDC.

Glutathione is the major low molecular weight thiol compound of the living plant or animal cell. It is a tripeptide with a gamma peptide linkage between the amine group of cysteine (which is attached by normal peptide linkage to a glycine) and the carboxyl group of the glutamate side-chain. It is an antioxidant, preventing damage to important cellular components caused by reactive oxygen species such as free radicals and peroxides. The sulfhydryl (thiol) group (SH) of cysteine serves as a proton donor and is responsible for the biological activity of glutathione. Glutathione suppresses human immunodeficiency virus expression in chronically infected monocytic cells. It is a useful tripeptide involved in many aspects of metabolism, including transport of g-glutanyl amino acids and reductive cleavage of disulfide bonds. Endogenous antioxidant that plays a major role in reducing reactive oxygen species formed during cellular metabolism and the respiratory burst. Glutathione-S-transferase catalyzes the formation of glutathione thioethers with xenobiotics, leukotrienes, and other molecules that have an electrophilic center. Glutathione also forms disulfide bonds with cysteine residues in proteins. Via these mechanisms, it can have the paradoxical effect of reducing the efficacy of anti-cancer agents.

In aqueous solutions, NHS ester hydrolytic degradation is a competing reaction whose rate increases with pH. The maleimide group is more stable than the NHS-ester group, but will slowly hydrolyze and lose its reaction specificity for sulfhydryls at pH values > 7.5. For these reasons, conjugations with these crosslinkers are usually performed at pH 7.2–7.5, with the NHS ester (amine-targeted) reacted before or simultaneous with the maleimide (sulfhydryl-targeted) reaction.
