Gravin (also known as A Kinase Anchor Protein 12, AKAP12) was cloned from a cDNA expression library screened with serum, from a myasthenia gravis patient. to Gravin were found in 22 out of 72 myasthemia gravis patients and absent in sera of normal individuals. This protein is a myristoylated high molecular weight cytoplasmic protein expressed in endothelial cells, cultured fibroblasts and osteosarcoma cells and highly expressed in the brain. Gravin can bind kinases like PKA and PKC by different domains of the protein and can localize these kinases to specific domains in the cell. Upon differentiation of NT2 cells to neurons (NT2 N), Gravin expression is up regulated and enriched at the inner peripheral cortex in close proximity to the plasma membrane. In differentiated cells, gravin is co-localized with PKCbII, PKCa and PKA. Decreased binding of PKCa to gravin was found after activation of PKC.

Modulation of the chromatin structure plays an important role in the regulation of transcription in eukaryotes. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

Stanniocalcin 1 (STC1) and stanniocalcin 2 (STC2) are mammalian peptide hormones that were previously considered to be present only in bony fish, where they are involved in calcium homeostasis. STC1 plays a role in calcium and phosphate homoeostasis and is phosphorylated in vitro by protein kinase C, and STC2 is phosphorylated in vitro by casein kinase II (CK2). A human fibrosarcoma cell line, HT1080, expresses both STC1 and STC2 as secreted phosphoproteins in vivo, with STC2 being phosphorylated by an ecto-CK2-like enzyme. STC1 and STC2 have opposite effects on calcium and phosphate homeostasis, namely anti-hypercalcemic and anti-hypocalcemic actions, respectively. STC1 and STC2 are detected in human adrenal tumours, such as pheochromocytoma, differentiated neuroblastoma aldosterone-producing adenoma, and in cultured adrenal tumour cells (rat pheochromocytoma PC-12 cells and human neuroblastoma NB-1 cells).

Localized to the nucleus, ZNF426 contains one KRAB domain and 12 C2H2-type zinc fingers through which it may convey DNA, RNA and protein binding capabilities. Specifically, ZNF426 may interact with the viral protein KSHV ORF 50 and, through this interaction, may activate viral gene transcription.

Stat1 and Stat2 are induced by IFN-å and form a heterodimer which is part of the ISGF3 transcription factor complex. Although early reports indicate Stat3 activation by EGF and IL-6, it has been shown that Stat3∫ appears to be activated by both while Stat3å is activated by EGF, but not by IL-6. Highest expression of Stat4 is seen in testis and myeloid cells. IL-12 has been identified as an activator of Stat4. Stat5 has been shown to be activated by Prolactin and by IL-3. Stat6 is involved in IL-4 activated signaling pathways.
