Serpin F1 is a secreted glycoprotein that belongs to the noninhibitory serpin. It has an alpha/beta core serine-protease inhibitor domain, three major beta-sheets, and ten alpha-helices. As protease inhibitors, serpins have an array of functions including regulating blood clotting, the complement pathway, extracellular matrix remodeling, and cell motility. They are also involved in activities that extend beyond their ability to inhibit proteases. For instance, they may also regulate blood pressure, angiogenesis, or act as storage/transport proteins. Serpin F1 is a new promising approach for the treatment of osteosarcoma and has been described as a natural angiogenesis inhibitor with neurotrophic and immune-modulation properties. The human serpin superfamily consists of at least 35 members that target not only serine proteases, but also selected cysteine proteases and non-protease proteins. Levels of the natural ocular anti-angiogenic factor SentrinF1 (PEDF) is associated with proliferative retinopathy.

LFG Antibody: Programmed cell death regulates a number of biological processes such as normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. LFG is a recently identified protein that can inhibit the apoptotic signal transduced by the Fas receptor but not from the related tumor necrosis factor-alpha death signal. In this respect, LFG is functionally similar to the anti-apoptotic proteins FAIM, FLIP and Bcl-xL. LFG, a seven membrane spanning protein, can bind the Fas receptor but does not regulate Fas expression or inhibit binding of FADD to Fas. LFG is widely distributed, but highly expressed in the hippocampus and other neural tissues. LFG was also identified as the neural membrane protein 35 (NMP35) and its expression is known to be regulated by the Phosphatidylinositol 3-kinase-Akt/PKB pathway.

Cytotoxic Tlymphocyte 4(CTLA-4,CD152), is a type I transmembrane T cell inhibitory molecule that is a member of the Ig superfamily. Human or mouse CTLA4 cDNA encodes 223 amino acids (aa) including a 35 aa signal sequence, a 126 aa extracellular domain (ECD) with one Ig-like V-type domain, a 21 aa transmembrane (TM) sequence, and a 41 aa cytoplasmic sequence. It is widely expressed with highest levels in lymphoid tissues. CD28 and CTLA-4, together with their ligands, B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. CTLA4 transmits an inhibitory signal to T cells, whereas CD28 transmits a stimulatory signal. Intracellular CTLA4 is also found in regulatory T Cells and may play an important role in their functions. Tcell activation through the Tcell receptor and CD28 leads to increased expression of CTLA4.

Serpin A5 is a member of the human Serpin superfamily consists of at least 35 members. It is synthesized in the liver and has been detected in saliva, cerebral spinal fluid, amniotic fluid, tears and semen. As a potent inhibitor of the protein C anticoagulant pathway at the levels of both zymogen activation and enzyme inhibition, Serpin A5 additionally inhibits a variety of serine protease including thrombin, factor Xa, several kallekreins and acrosin. It plays a critical role in the processes of blood of blood coagulation and fertilization. Serpin A5 also inhibits urinary plasminogen activator (uPA), a mediator of tumor cell invasion, and regulates tumor growth and metastasis by inhibiting angiogenesis. Furthermore, recent studies have identified PCI as a potent and direct inhibitor of activated HGFA (hepatocyte growth factor activator), suggesting a novel function in the regulation of tissue repair and regeneration. Similar to Serpins C1 and D1, the thrombin inhibitory activity of serpinA5 is enhanced by heparin.

Some of these IL-12 activities include the induction of IFN-gamma and TNF in resting and activated T and NK cells; the enhancement of cytotoxic activity of resting NK and T cells, the stimulation of resting T cell proliferation in the presence of a comitogen; and the enhancement of NK cell proliferation. Current evidence indicates that IL-12 is a key mediator of cellular immunity and induces the differentiation of Th1 cells from precursor T helper cells. Based on its activities, it has been suggested that IL-12 may have therapeutic potential as a vaccine adjuvant that promotes cellular immunity and as an antitumour and anti viral agent.
