Connexin 29 belongs to the connexin family and is a member of the epsilon-type subfamily. Connexin 29 is a membrane bound, multi-pass protein also known as gap junction epsilon-1 protein. A connexon, consisting of connexin hexamers, is a membrane bound structure that is integral in the formation of a gap junction. One gap junction consists of a cluster of closely packed pairs of transmembrane channels, the connexons, through which materials of low molecular weight diffuse from one cell to a neighboring cell. Connexin 29 expression is restricted to the central nervous system and is present in brain, spinal cord, and sciatic nerve samples. It has been suggested that connexin 29 in the mature CNS contributes minimally to gap junctional intercellular communication in oligodendrocyte cell bodies. Rather, connexin 29 is targeted to myelin where it, along with connexin 32, may contribute to connexin-mediated communication between adjacent layers of uncompacted myelin.

The oligodendrocyte lineage-specific basic helix-loop-helix (OLIG) family of transcription factors include OLIG1-OLIG3, which differ in tissue expression. OLIG1 and OLIG2 are specifically expressed in nervous tissue as gene regulators of oligodendrogenesis. OLIG2 is more widely expressed in embryonic brain than OLIG1, while OLIG3 is primarily expressed in non-neural tissues. OLIG1 and OLIG2 interact with the Nkx-2.2 homeodomain protein, which is responsible for directing ventral neuronal patterning in response to graded Sonic hedgehog signaling in the embryonic neural tube. These interactions between OLIG proteins and Nkx-2.2 appear to promote the formation of alternate cell types by inhibiting V3 interneuron development. OLIG1 and OLIG2 are abundantly expressed in oligodendroglioma and nearly absent in astrocytomas. Therefore, OLIG proteins are candidates for molecular markers of human glial brain tumors, which are the most common primary malignancies of the human brain.

Histones are highly conserved proteins that serve as the structural scaffold for the organization of nuclear DNA into chromatin. The four core histones, H2A, H2B, H3, and H4, assemble into an octamer (2 molecules of each). Subsequently, 146 base pairs of DNA are wrapped around the octamer, forming a nucleosome, the basic subunit of chromatin. Histone modifications regulate DNA transcription, repair, recombination, and replication. The most commonly studied modifications are acetylation, phosphorylation, methylation, and ubiquitination. These modifications can alter local chromatin architecture, or recruit trans-acting factors that recognize specific histone modifications (the "histone code" hypothesis). Trimethylation of histone H3 on Lys9 (H3K9me3) is one of the most highly studied epigenetic marks. H3K9me3 functions in the repression of euchromatic genes, and in epigenetic control of heterochromatin assembly, most likely via acting as a recognition motif for the binding of chromatin-associated proteins, such as Swi6 or HP1Alpha/Beta. The enzymes responsible for H3K9me3 formation are SUV39H1 and SUV39H2.

Required for V(D)J recombination, the process by which exons encoding the antigen-binding domains of immunoglobulins and T-cell receptor proteins are assembled from individual V, (D), and J gene segments. V(D)J recombination is initiated by the lymphoid specific RAG endonuclease complex, which generates site specific DNA double strand breaks (DSBs). These DSBs present two types of DNA end structures: hairpin sealed coding ends and phosphorylated blunt signal ends. These ends are independently repaired by the non homologous end joining (NHEJ) pathway to form coding and signal joints respectively. This protein exhibits single-strand specific 5'-3' exonuclease activity in isolation and acquires endonucleolytic activity on 5' and 3' hairpins and overhangs when in a complex with PRKDC. The latter activity is required specifically for the resolution of closed hairpins prior to the formation of the coding joint. May also be required for the repair of complex DSBs induced by ionizing radiation, which require substantial end-processing prior to religation by NHEJ.

Crk p38 (CrkII) is an isoform of the protein Crk. CrkI and CrkII are produced from the same crk gene by alternative splicing. The two isoforms differ in their biological activities with CrkII having less transforming activity than CrkI.Crk p38 is a member of an adapter protein family that binds to several tyrosine-phosphorylated proteins. It has several SH2 and SH3 domains (src-homology domains) and is involved in several signaling pathways, recruiting cytoplasmic proteins in the vicinity of tyrosine kinase through SH2-phosphotyrosine interaction. The N-terminal SH2 domain of this protein functions as a positive regulator of transformation whereas the C-terminal SH3 domain functions as a negative regulator of transformation. Crk is believed to be a regulator of invasive responses because increased levels of the protein have been observed in multiple human cancers. Ficolin B is the designation in mouse and rat of a protein also known as L-ficolin, ficolin-2, collagen/fibrinogen domain-containing protein 2, serum lectin p35, EBP-37 or hucolin. Ficolin B is a 313 amino acid member of the ficolin lectin family. It is a secreted innate immunity pattern recognition molecule expressed in liver and plasma that forms a disulfide-linked homopolymer. This extensive N-terminal disulfide bridge formation can lead to a functional dodecamer polypeptide. Ficolin B binds to DNA ligands expressed by late apoptotic and necrotic cells to increase attachment and engulfment. Variation in ficolin B concentrations amongst individuals is associated with polymorphisms in the promoter and structural portion of the FCN2 gene. In patients with Behé´t's disease (BD), there exists a significant difference in allele frequency for FCN2 gene single nucleotide polymorphisms (SNPs) within the -557 and -64 promoter sites within HLA-B51 positive and HLA-B51 negative subgroups.
