Vascular endothelial growth factor (VEGF) is a major growth factor for endothelial cells. This gene encodes one of the two receptors of the VEGF. This receptor, known as kinase insert domain receptor, is a type III receptor tyrosine kinase. It functions as the main mediator of VEGF-induced endothelial proliferation, survival, migration, tubular morphogenesis and sprouting. The signalling and trafficking of this receptor are regulated by multiple factors, including Rab GTPase, P2Y purine nucleotide receptor, integrin alphaVbeta3, T-cell protein tyrosine phosphatase, etc.. The autosomal dominant cerebellar ataxias (ADCA) are a heterogeneous group of neurodegenerative disorders characterized by progressive degeneration of the cerebellum, brain stem and spinal cord. Clinically, ADCA has been divided into three groups: ADCA types I-III. ATX2 belongs to the autosomal dominant cerebellar ataxias type I (ADCA I) which are characterized by cerebellar ataxia in combination with additional clinical features like optic atrophy, ophthalmoplegia, bulbar and extrapyramidal signs, peripheral neuropathy and dementia. ATX2 is caused by expansion of a CAG repeat in the coding region of ATX2. Longer expansions result in earlier onset of the disease. There are four named isoforms.

FNDC1, also known as MEL4B3, Ags8, Expressed in synovial lining protein and Activation-associated cDNA protein, is a 1888 amino acid secreted protein that contains five fibronectin type-III domains. FNDC1 is moderately expressed in skeletal muscle, pancreas, heart, kidney, spinal cord, ovary and lung. Expression of FNDC1 is induced in response to hypoxia in ventricular cardiomyocytes. Since FNDC1 interacts with G∫ and G©, it is likely that FNDC1 is an activator for G-protein signaling. Though normally absent in healthy skin, FNDC1 expression is induced by TGF-beta signaling in skin tumors and psoriasis. There are two isoforms of FNDC1 that are produced as a result of alternative splicing events.

Eukaryotic elongation factor 2 kinase (EEF2k) previously known as Ca2+/calmodulin dependent protein kinase III, is an abundant cytoplasmic protein highly specific for elongation factor 2 (eEf2). Phosphorylation of eEF2 by eEF2 kinase on specific threonine residues results in the inactivation of eEF-2 and in termination of mRNA translation. The activity of eEF2 kinase is not only dependent upon Ca2+ ions, calmodulin (CaM) and insulin, but is also regulated both negatively and positively via phosphorylation by different protein kinases (AMPK, S6K1, p90 RSK). There is also evidence that eEF-2 phosphorylation is involved in the regulation of cell cycle progression, cellular differentiation, oogensis and malignant tumors.

FNDC3A is a 1,134 amino acid protein that belongs to the FNDC3 family of proteins. FNDC3A contains an N-terminal proline-rich region, nine fibronectin type-III domains (none of which contain an RGD sequence) and a hydrophobic C-terminal transmembranous helix. Expressed in a wide variety of tissues, FNDC3A localizes to Golgi vesicles and to the developing acrosome of spermatids. FNDC3A is believed to function in glycosaminoglycan and collagen synthesis. In mice, a mutation in the gene encoding FNDC3A causes male sterility due to defective adhesion between Sertoli cells and spermatids in the seminiferous epithelium. This suggests that FNDC3A plays an important role in spermatogenesis, possibly mediating or maintaining the adhesion between Sertoli cells and spermatids.
