Beschreibung:
FNDC5 is a membrane protein comprised by a short cytoplasmic domain, a transmembrane segment, and an ectodomain consisting of a fibronectin type III (FNIII) domain. The ectodomain has been proposed to be cleaved to a soluble peptide hormone named “irisin”. Irisin is secreted from muscle in response to exercise, and may regulate energy metabolism, stem cell differentiation, and neuronal development. The circulating irisin converts white fat into the more thermogenic beige fat, and may mediate some beneficial effects of exercise in humans and the potential for generating weight loss. In mice, expression of Irisin has been shown to regulate obesity and diabetes. A similar function is suggested in human.

Voltage-gated K+ channels are important determinants of neuronal membrane excitability. Moreover, differences in K+ channel expression patterns and densities contribute to the variations in action potential waveforms and repetitive firing patterns evident in different neuronal cell types (Maletic-Savatic et al., 1995; Pongs, 1999; Blaine and Ribera, 1998; Burger and Ribera, 1996). The Kv3.1 potassium channel is expressed at high levels in neurons that characteristically fire rapid trains of action potentials (Gan et al., 1999). Particularly high levels of this channel are found in neurons of the auditory brainstem. These neurons appear to participate in neural circuits that determine the intensity and timing of auditory stimuli and use this information to determine the location of sounds in space (von Hehn et al., 2004).

Phosphatidylinositol 4-Kinase beta (PI4KB) belongs to the PI3/PI4-kinase family and Type III PI4K subfamily. PI4KB contains one PI3K/PI4K domain and one PIK helical domain. It is highly expressed in heart, skeletal muscle, pancreas, testis and ovary,; it is lowly expressed in liver tissue. PI4KB kinase activity can be inhibited by wortmannin, and increased by interacting with NCS1/FREQ. PI4KB phosphorylates phosphatidylinositol (PI) in the first committed step in the production of the second messenger inositol-1,4,5,-trisphosphate (PIP). In addition, PI4KB may regulate Golgi disintegration/reorganization during mitosis.

Tumour necrosis factor receptor superfamily member 27, also known as XEDAR and EDA2R, is a type III transmembrane protein of the TNFR (tumour necrosis factor receptor) superfamily, and contains 3 cysteine-rich repeats and a single transmembrane domain but lacks an N-terminal signal peptide. EDA2R, as well as its paralog, EDAR, binds the ectodysplasin ligands EDA-A2 and EDA-A1, which are two alternatively spliced forms of the EDA gene. Mutations in the EDA gene are associated with the X-linked form of Hypo hidrotic Ectodermal Dysplasia (HED), a disease typically characterized by abnormal hair, teeth and sweat glands.

neuregulin-1 (heregulin-1，NRG1) is a member of neuregulin family, which is comprised of four genes that encode a large number of secreted or membrane-bound isoforms. The TEK receptor tyrosine kinase is expressed almost exclusively in endothelial cells in mice, rats, and humans. This receptor possesses a unique extracellular domain containing 2 immunoglobulin-like loops separated by 3 epidermal growth factor-like repeats that are connected to 3 fibronectin type III-like repeats. The ligand for the receptor is angiopoietin-1. Defects in TEK are associated with inherited venous malformations; the TEK signaling pathway appears to be critical for endothelial cell-smooth muscle cell communication in venous morphogenesis. TEK is closely related to the TIE receptor tyrosine kinase.
