Recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.12. Workshop IV; Code B17). It recognizes an extracellular domain of CD20. It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. It acts as calcium channel involved in B cell activation and cell cycle progression.

Syntaxins were originally thought to be docking proteins, but have more recently been categorized as anchoring proteins that anchor themselves to the cytoplasmic surfaces of cellular membranes. Syntaxins bind to various proteins involved in exocytosis, including VAMPs (vesicle-associated membrane proteins), NSF (N-ethylmaleimide-sensitive factor), SNAPs (soluble NSF attachment proteins) and Synaptotagmin. Endobrevin, also designated VAMP-8 or ED, is a 100 amino acid single-pass type IV membrane protein that belongs to the synaptobrevin family. Similar in sequence to the synaptobrevins, endobrevin is abundantly expressed in kidney, moderately expressed in heart and spleen, and slightly expressed in brain, thymus and liver. Endobrevin interacts specifically with the SNAPs, most likely through an endobrevin-containing SNARE complex.

Defects in G6PD are the cause of chronic non-spherocytic hemolytic anemia (CNSHA) . Deficiency of G6PD is associated with hemolytic anemia in two different situations. First, in areas in which malaria has been endemic, G6PD-deficiency alleles have reached high frequencies (1% to 50%) and deficient individuals, though essentially asymptomatic in the steady state, have a high risk of acute hemolytic attacks. Secondly, sporadic cases of G6PD deficiency occur at a very low frequencies, and they usually present a more severe phenotype. Several types of CNSHA are recognized. Class-I variants are associated with severe NSHA; class-II have an activity <10% of normal; class-III have an activity of 10% to 60% of normal; class-IV have near normal activity.

BM88 is a 149 amino acid protein that belongs to the CEND1 familly. Involved in neuroblastoma cell differentiation, BM88 is a single-pass type IV membrane protein that is neuron specific. It is suggested that BM88 forms a dimer of two identical polypeptides linked by disulfide bridges. BM88 has a central proline-rich region containing four PxxP motifs, which typically bind SRC homology-3 (SH3) domains, as well as a putative C-terminal transmembrane region, and several potential sites for N-glycosylation, myristoylation and phosphorylation. It is also suggested that a novel signaling mechanism exists by which BM88 interferes with calcium release from inositol 1,4,5-trisphosphate-sensitive stores and exerts anti-proliferative and anti-apoptotic functions. BM88 is an important molecular target for HDAC inhibition, and transcription of BM88 is induced by trichostatin-A.

Recognizes a protein of 40 kDa, identified as CD7 (Workshop IV; Code T155). CD7 is a member of the immunoglobulin gene superfamily. Its N-terminal amino acids 1-107 are highly homologous to Ig kappa-L chains whereas the carboxyl-terminal region of the extracellular domain is proline-rich and has been postulated to form a stalk from which the Ig domain projects. CD7 is expressed on the majority of immature and mature T-lymphocytes, and T cell leukemia. It is also found on natural killer cells, a small subpopulation of normal B cells and on malignant B cells. Cross-linking surface CD7 positively modulates T cell and NK cell activity as measured by calcium fluxes, expression of adhesion molecules, cytokine secretion and proliferation. CD7 associates directly with phosphoinositol 3'-kinase. CD7 ligation induces production of D-3 phosphoinositides and tyrosine phosphorylation.

Matrix metalloproteinase-7 (MMP-7) also known as matrilysin and PUMP (EC 3.4.24.23) cleaves a number of substrates including collagen types IV and X, elastin, fibronectin, gelatin, laminin and proteoglycans. MMP-7 is closely related to the stromelysin family members but is encoded by a different gene. MMP-7 is the smallest of all the MMPs consisting of a pro-peptide domain and a catalytic domain. It lacks the hemopexin-like domain common to other members of the MMPs. MMP-7 is secreted as a 28kD proenzyme and can be activated in vitro by organomercurials and trypsin and in vivo by MMP-3 to a 18kD active MMP-7 enzyme. Once activated, MMP-7 can activate pro-MMP-1 and pro-MMP-9 but not pro-MMP-2. MMP-7 is widely expressed having been reported in elevated levels in cycling endometrium as well as in colorectal cancers and adenomas, hepatocellular carcinomas, rectal carcinomas, and approximately 50% of gliomas.

The kinesin superfamily of proteins (KIFs) consists of a class of microtubule-dependent motors that play a major role in many cellular and developmental functions, including organelle transport, mitosis, meiosis, and possibly long-range signaling in neurons. The kinesin proteins are involved in organelle transport and are primarily associated with anterograde transport of vesicles and organelles in neurons, epithelial cells, and melanosomes with bidirectional transport of mitochondria. They also mediate transport between the endoplasmic reticulum (ER) and the Golgi complex. In neurons, kinesin motors conduct vesicular transport, such as of synaptic vesicle components to axons and of neurotransmitter receptors to dendrites. KIF17 belongs to the functionally diverse subgroup of the kinesin superfamily characterized by a N-terminal motor domain (N-IV class), that includes the KIF3 motor protein. KIF17 is specifically expressed in the brain, present in abundance in the gray matter, particularly in the hippocampus and cerebral cortex, but not in the white matter such as the optic nerve.
