Voltage-modulated Ca(2+)-activated, monovalent cation channel (VCAM) that mediates a transient membrane depolarization and plays a central role in taste transduction. Monovalent-specific, non-selective cation channel that mediates the transport of Na(+), K(+) and Cs(+) ions equally well. Activated directly by increases in intracellular Ca(2+), but is impermeable to it. Gating is voltage-dependent and displays rapid activation and deactivation kinetics upon channel stimulation even during sustained elevations in Ca(2+). Also activated by a fast intracellular Ca(2+) increase in response to inositol 1,4,5-triphosphate-producing receptor agonists. The channel is blocked by extracellular acidification. External acidification has 2 effects, a fast reversible block of the current and a slower irreversible enhancement of current inactivation. Is a highly temperature-sensitive, heat activated channel showing a steep increase of inward currents at temperatures between 15 and 35 degrees Celsius. Heat activation is due to a shift of the voltage-dependent activation curve to negative potentials. Activated by arachidonic acid in vitro. May be involved in perception of bitter, sweet and umami tastes. May also be involved in sensing semiochemicals.

EHEC kann lebensbedrohliche Krankheiten auslösen, insbesondere bei Menschen mit Immunschwäche, kleinen Kindern und älteren Menschen. Obwohl die häufigste Ursache von EHEC <i> ist E. coli O157, andere Serotypen wie O26, O45, O103, O111, O121 und O145 sind ebenfalls relativ häufige Infektionsursachen. Typische Symptome sind starke Bauchschmerzen und Durchfall, der zunächst wässrig, aber oft blutig wird. E. coli</i> sind gramnegative, nicht sporulierende, fakultativ anaerobe Bazillen. Unter den pathogenen <i>E. coli</i> haben in den letzten Jahren Verotoxin-bildende Stämme (STEC oder VTEC) an Bedeutung gewonnen. Die Gruppe der enterohämorrhagischen <i>E. coli</i> (EHEC) mit seinem hochpathogenen Serovar O157:H7-Stamm ist in dieser Hinsicht besonders interessant. Hauptansteckungsquellen sind kontaminierte, rohe oder unzureichend erhitzte Lebensmittel tierischen Ursprungs, wie Fleisch und Milchprodukte. Das Reservoir für EHEC ist der Darm von Wiederkäuern. Bei der Verarbeitung von Fleisch- und Milchprodukten können Mikroorganismen bei unzureichenden hygienischen Bedingungen in Lebensmittel gelangen. Der drastische Anstieg der Lebensmittelkontamination durch <i>E. coli</i> O157 erfordert zuverlässige und schnelle Nachweismethoden. Innovative Schnelltests erleichtern das Screening auf Krankheitserreger. Komplette Test-Sets zum Nachweis von Krankheitserregern und Toxinen. Das einstufige Format reduziert die Möglichkeit von Fehlern bei der Handhabung. CCT2 is one of eight largely unrelated subunit proteins found in a protein chaperone complex known as the chaperonin-containing TCP-1 (CCT) or TRiC complex. The CCT complex is an abundanct cytoslic component that is credited with helping newly synthesized polypeptides adopt the correct conformation (1). Proteins that fold and assemble with the help of CCT include the cytoskeletal proteins actin and tubulin as well as up to 15% of newly synthesized eukaryotic proteins (2). CCT2 is the β-subunit of the chaperone complex and is one of several CCT proteins that exhibit increased expression in response to stress. This implies that the CCT complex helps cells recover from protein damage by assisting in protein folding and assembly (3). CCT subunit levels also change throughout the cell cycle, with lower proteins levels (and reduced chaperone activity) found during induced cell cycle arrest during at M phase (4). Each CCT subunit is thought to perform a specific function during protein folding and assembly (5); CCT2 exhibits both actin and tubulin binding activities (6,3) but the exact molecular function on this subunit remains uncertain.

Two splice variants with different carboxyterminal ends were described. The message was found only in the brain in this publication. Another group identified ADAM11 in the human brain, where ADAM11 was thought to be involved in cell migration and spatial patterning. ADAM11 was mapped to 17q21.3, a region of interest for breast cancer, and mutations in ADAM11 are associated with some breast cancers. Retinoic acid caused a doubling in ADAM11 message levels over 24 hours in NT2/D1 cells, a human embryonic carcinoma cell line. ADAM11 null mutant mice have deficits in spatial learning and motor coordination, although they did have normal cell migration and differentiation during development. ADAM11 is a member of the ADAMs family (A Disintegrin And Metalloproteinase), but does not contain the canonical HExxHxxxxH zinc-binding metalloproteinase catalytic site. The domain structure of the full-length ADAM11 includes a signal sequence, propeptide domain, metalloproteinase-like domain, disintegrin-like domain, cys-rich domain, EGF-like domain, a spacer region, then the transmembrane domain and a short cytoplasmic domain.
