This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. Expressed primarily in brain, HS1BP3 is encoded by a gene mapping to human chromosome 2p24.1. The gene encoding HS1BP3 is frequently mutated in familial essential tremor, a disorder characterized by kinetic tremor the the hands, voice or head, though there is no correlation to Parkinson disease. HS1BP3 interacts with HAX-1’s SH3 domain, and may also play a role in the regulation of catecholamine and serotonin metabolism. Acting as a regulator of IL-2 signaling, HS1BP3 is likely involved in lymphocyte activation.

CRTAC1 is a 661 amino acid secreted protein. CRTAC1 is O-glycosylated and contains one EGF-like domain and four FG-GAP repeats. Up-regulated in mesenchymal stem cells undergoing chondrogenic differentiation and also by growth factor BMP-4, CRTAC1 is found in the interterritorial matrix of articular deep zone cartilage. CRTAC1 is expressed as three isoforms produced by alternative splicing events. All isoforms are expressed in bone, cartilage and lung, while isoforms 1 and 2 are expressed in the brain. The gene that encodes CRTAC1 maps to human chromosome 10, which contains over 800 genes and 135 million nucleotides, making up nearly 4.5% of the human genome.

The PSD-95/SAP 90 family of proteins, which are known to bind to and cluster various membrane proteins, are involved in the organization of synaptic structure. SynGAP (for synaptic GTPase activating protein), a novel synaptic RasGAP, is a brain-specific protein abundant in the cortex, hippocampus and olfactory bulb. SynGAP interacts with all three PDZ domains within PSD-95/ SAP 90 proteins. It represents one of three classes of mammalian RasGAPs and is specifically localized to excitatory synapses possessing NMDA receptors. SynGAP may be involved in the regulation of BDNF as well as Ras signaling. Its activity is inhibited by phosphorylation by CaMKII, which could result in the activation of the MAP kinase pathway.

Mutations in the CLN8 gene cause neuronal ceroid lipofuscinosis 8 and progressive epilepsy with mental retardation (EPMR). Both disorders are forms of neuronal ceroid-lipofuscinose (NCL), a group of progressive neurodegenerative diseases found in children, characterised by failure of psychomotor development, impaired vision, seizures and premature death. The CLN8 protein is one of eight proteins in the CLN family, including CLN1-CLN7, which are associated with NCL.

Expressed in a wide variety of tissues, C1orf25 contains one C2H2-type zinc finger and shares sequence similarity with TRMT1, a tRNA methyltransferase that dimethylates guanine at position 26 of tRNAs. As is suggested by observations of C1orf25-deficient mice, C1orf25 is believed to play a role in exploratory behavior and motor coordination. MCM3 is one of the highly conserved mini-chromosome maintenance proteins that are involved in the initiation of eukaryotic genome replication. The hexameric protein complex formed by MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. This protein is a subunit of the protein complex that consists of MCM2-7. It has been shown to interact directly with MCM5/CDC46. This protein also interacts with, and thus is acetlyated by MCM3AP, a chromatin-associated acetyltransferase. The acetylation of this protein inhibits the initiation of DNA replication and cell cycle progression.
