Mutations in ZMPSTE24 gene have been associated with mandibuloacral dysplasia and restrictive dermopathy.The protein encoded by this gene is a zinc metalloprotease similar to yeast Ste24p. It is an integral membrane protein belonging to peptidase family M48 and is found in the endoplasmic reticulum and possibly in the Golgi compartment. It is thought to be involved in the proteolytic processing of farnesylated proteins.

SG-Glutamic-C™ is a serine endopeptidase, from<i> S. aureus V8</i>, that is highly specific for the cleavage of peptide bonds at the carboxy side of either aspartic or glutamic acid, depending on the buffer used. In Tris-HCl buffer, in particular in the absence of phosphate ions, the enzyme is specific for the glutamyl site. Recommended buffers for fragmentation of proteins using this enzyme are 50 mM Tris-HCI, pH 8,0 or bicarbonate buffer. Highly purified preparations of SG-Glutamic-C™ are chemically modified making the enzyme both resistant to autolysis and stabilises its enzymatic activity.

Splicing factor 3b is also a component of the minor U12-type spliceosome. The carboxy-terminal two-thirds of subunit 1 have 22 non-identical, tandem HEAT repeats that form rod-like, helical structures.This gene encodes subunit 1 of the splicing factor 3b protein complex. MIPU1 Antibody: Mipu1, also known as zinc finger protein 667 or ZNF667, encodes a nuclear-localized protein containing 14 carboxy-terminal zinc finger motifs and an amino-terminal KRAB domain. This protein is highly expressed in heart and brain and is upregulated in rat heart after a transient ischemia-reperfusion procedure. Overexpression experiments suggest that Mipu1 suppresses the transcriptional activities of AP-1 and SRE in the MAPK signaling pathway and thus may play a role in the pathogenesis of cardiac and vascular disease. SIPA1L1 Antibody: Signal-induced proliferation associated-like protein 1 (SIPA1L1) is a member of the SIPA1 family of RapGAPs. SIPA1L1 was initially identified as a binding partner and degradation target of the E6 oncoprotein of high-risk papillomaviruses. Recently, it was discovered that Casein kinase I epsilon (CKIe), a Wnt-regulated kinase that regulates Wnt/b-catenin signaling, also can bind to the carboxy-terminus of SIPA1L1. CKIe phosphorylates SIPA1L1, thereby reducing its stability and alleviating its inhibition of Rap1, a protein required for Wnt8/CKIe-mediated gastrulation during embryogenesis, suggesting SIPA1L1 plays important roles in embryo development as well as control of cell proliferation.

Vinculin is a cytoskeletal protein that plays an important role in the regulation of focal adhesions and embryonic development. Three structural vinculin domains include an amino-terminal head, a short flexible proline-rich region and a carboxy-terminal tail. Expression of vinculin were shown to be affected by the level of actin expression. Vinculin deficiencies are associated with a decrease in cell adhesion and an increase in cell motility, suggesting a possible role in metastatic growth. Defects in VCL are the cause of cardiomyopathy dilated type 1W (CMD1W).

TMEM16B Antibody: Calcium-activated chloride channels (CaCC) are present in many cell types and mediate physiological functions such as epithelial secretion, sensory signal transduction, and smooth muscle contraction. Subunits of these CaCC's include the transmembrane proteins TMEM16A and TMEM16B. TMEM16B is predicted to have eight transmembrane domains with both the amino and carboxy termini in the cytoplasm and is expressed in several tissues including olfactory sensory neurons as well as photoreceptors in mammalian retina. Like TMEM16A, TMEM16B is thought to form at least part of CaCC's but has different biophysical characteristics such as voltage dependence and unitary conductance.
