The tetratricopeptide repeat (TPR) motif is a degenerate, 34 amino acid sequence found in many proteins and acts to mediate protein-protein interactions in various pathways. At the sequence level, there can be up to 16 tandem TPR repeats, each of which has a helix-turn-helix shape that stacks on other TPR repeats to achieve ligand binding specificity. EDRF1 (erythroid differentiation-related factor 1), also known as C10orf137 (chromosome 10 open reading frame 137), is a 1,238 amino acid protein containing two TPR repeats. Localizing to nucleus, EDRF1 is involved in transcriptional activation of globin genes by regulating DNA-binding activity of GATA-1 transcription factor. EDRF1 may also play an important role in organ development and histological differentiation. EDRF1 exists as four alternatively spliced isoforms and is encoded by a gene mapping to human chromosome 10q26.13.

Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDHB12 (Protocadherin beta 12) is a 795 amino acid protein that is one of 16 proteins in the protocadherin beta cluster. Unlike the alpha and gamma gene clusters whose genes are spliced to downstream constant region exons during transcription, members of the beta cluster (such as PCDHB12) do not use constant-region exons to produce mRNAs. As a result, each protocadherin beta gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. Localized to the cell membrane, PCDHB12 is a single-pass type I membrane protein that contains six cadherin domains. PCDHB12 may participate in the establishment and maintenance of specific neuronal connections in the brain.

Semaphorins are a family of cell surface and secreted proteins involved in neural development that are conserved from insects to humans. Members of this family are approximately 750 amino acids in length (including signal sequences) and are defined by a conserved extracellular semaphorin domain of approximately 500 amino acids containing 14-16 cysteines, blocks of conserved sequences and no obvious repeats. The transmembrane semaphorins are characterised by an additional 80 amino acid transmembrane domain and an 80-110 amino acid cytoplasmic domain. Secreted and cell-bound semaphorins chemically attract and repel the growth of neural axons, guiding the development of intricate networks of neural tissue. SEMA3E is a secreted semaphorin with 775 amino acids. Mutations in the SEMA3E gene are associated with CHARGE syndrome, a disorder characterised by cranial nerve dysfunction, coloboma of the eye, choanal atresia, inner and external ear abnormalities, cardiac anomalies, genitourinary abnormalities, and growth retardation.

Heterotrimeric G proteins function to relay information from cell surface receptors to intracellular effectors. Each of a very broad range of receptors specifically detects an extracellular stimulus (i.e. a photon, pheromone, odorant, hormone or neurotransmitter), while the effectors (e.g. adenyl cyclase), which act to generate one or more intracellular messengers, are less numerous. In mammals, G protein alhfa, beta and gamma polypeptides are encoded by at least 16, 4 and 7 genes, respectively. Most interest in G proteins has been focused on their a subunits, since these proteins bind and hydrolyze GTP and most obviously regulate the activity of the best studied effectors. Evidence, however, has established an important regulatory role for the beta gamma subunits. It is becoming increasingly clear that different G protein complexes expressed in different tissues carry structurally distinct members of the gamma as well as the alhfa and beta subunits, and that preferential associations between members of subunit families increase G protein functional diversity.

The semaphorins are a family of proteins that are involved in signaling. All the family members have a secretion signal, a 500-amino acid sema domain, and 16 conserved cysteine residues(Kolodkin et al., 1993 [PubMed 8269517]). Sequence comparisons have grouped the secreted semaphorins into 3 general classes, all of which also have an immunoglobulin domain. The semaphorin III family, consisting of human semaphorin III (SEMA3A; MIM 603961), chicken collapsin, and mouse semaphorins A, D, and E, all have a basic domain at the C terminus. Chicken collapsin contributes to path finding by axons during development by inhibiting extension of growth cones (Luo et al., 1993 [PubMed 8402908]) through an interaction with a collapsin response mediator protein of relative molecular mass 62K (CRMP62) (Goshima et al., 1995 [PubMed7637782]), a putative homolog of an axonal guidance associated UNC33 gene product (MIM 601168). SEMA3F is a secreted member of the semaphorin III family.

Interferon (IFN) Inducible 16 (IFI16) protein belongs to a family of HIN 200 human and mouse proteins. IFI16 is a nuclear protein comtaining regulatory domains such as DNA binding domain, transcriptional regulatory domain and DAPIN/PAAD domain. IFI16 has three isotypes A, B, and C (85-95 kDa), which arise as a result of mRNA alternative splicing. All are phosphorylated on serine and threonine residues and can homo and heterodimerize. Expression is restricted to the nuclei of hematopoietic cells, fibroblasts and epithelial cells. IFI16 expression in hematopoietic cells of myeloid lineage is tightly regulated and highly induced in the differentiation and proliferation of the cell. Due to its localisation in the nucleus, regulation of protein expression, and ability to bind DNA, it is assumed that IFI16 has a role in transcription regulation of cell differentiation. In addition, it was found that IFI16 can act as a transcriptional repressor and is involved in regulation and activation of p53 in cancer cells.
