ALKB protects DNA and RNA against damage from methylating compounds from the environment by directly reversing 1-methyladenine (1-meA) and 3-methylcytosine (3-meC) cytotoxic alkylation lesions in DNA and RNA. The enzymes act by oxidative demethylation, utilizing ferrous iron and alpha-ketoglutarate as cofactors, 2-oxoglutarate as a co-substrate, and molecular oxygen as the oxidizing agent. Deficiencies in DNA and RNA repair in mammals are associated with cancer, neurological disease and developmental defects. ALKB plays a role in resistance to anti-cancer drugs which attempt to damage tumour DNA. Escherichia coli ALKB protein belongs to the superfamily of 2-oxoglutarate- and iron(II)-dependent oxygenases.

This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained.Immunohistochemical staining with this MAb is indistinguishable from that obtained with monoclonal antibody 5D3.

The protein encoded by this gene is the 80-kilodalton subunit of the Ku heterodimer protein which is also known as ATP-dependant DNA helicase II or DNA repair protein XRCC5. Ku is the DNA-binding component of the DNA-dependent protein kinase, and it functions together with the DNA ligase IV-XRCC4 complex in the repair of DNA double-strand break by non-homologous end joining and the completion of V(D)J recombination events. This gene functionally complements Chinese hamster xrs-6, a mutant defective in DNA double-strand break repair and in ability to undergo V(D)J recombination. A rare microsatellite polymorphism in this gene is associated with cancer in patients of varying radiosensitivity.

GBGT1 is a 347 amino acid single-pass type II membrane protein that belongs to the glycosyltransferase 6 family. Localizing to the golgi apparatus membrane, GBGT1 is widely expressed, with high levels found in placenta, ovary and peripheral blood leukocyte, and lower levels expressed in liver, thymus and testis. GBGT1 utilizes manganese as a cofactor, and assists in the addition of N-acetylgalactosamine (GalNAc) in alpha-1,3-linkage to various substrates, resulting in the formation of glycolipids. Glycolipids are present in most eukaryotic cells and may assist in the adherence of certain pathogens. Existing as two alternatively spliced isoforms, the gene encoding GBGT1 maps to human chromosome 9q34.2 and mouse chromosome 2 A3.

However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Expression of CK10 is related to maturation of malignant keratinocytes, being preferentially detected in more-differentiated parts.

Encoding more than 700 genes, chromosome 15 is made up of approximately 106 million base pairs and comprises about 3% of the human genome. CREB3L3 is a 461 amino acid single-pass type II membrane protein that localizes to the endoplasmic reticulum (ER) and, in response to ER stress, is cleaved and translocated to the nucleus. Expressed exclusively in liver, CREB3L3 functions as a transcription factor that, during ER stress, is thought to activate genes that are involved in both the unfolded protein response and the acute phase response (APR). Additionally, CREB3L3 is underexpressed in hepatocellular carcinoma, suggesting a possible role as a tumor suppressor. CREB3L3 functions as a dimer and contains one leucine zipper domain, a KDEL-like sequence and a bZIP domain, through which it conveys its DNA binding ability. Three isoforms of CREB3L3 exist due to alternative splicing events.
